PartⅠ:preparation of porcine model of hepatic cirrhosis by gastrostomy tube assisted with diet controlobjective To explore the feasibility of preparation of porcine model of hepatic cirrhosis by gastrostomy tube assisted with diet control and understand the relationship between the cirrhosic level and some certain biochemical indexes,In order to offer a better animal model for observing surgical therapeutic efficacy.Methods Choosing the common pig as experimental animal,by temporary gastrostomy tube,CCl4 which is to be diluted with identical capacity of fat milk is administered according to 0.25ml/kg through T tube,then 30%edible alcohol containing 60mL is infused,twice a week;Their diet was simple Corn meal and 5%alcohol-water mixtures.during the course of the experimental procedure,we cut little pieces of the hepatic tissue which was prepared for HE stain and VG stain,collecting blood samples in order to test the indexes such as the function of liver and kidney,albumin,bilirubin and so on regularly.Results(1) According to Madden's staging for live cirrhosis,there are four porcines withⅢhepatic lesion and three porcines withⅡhepatic lesion and two porcines withⅣhepatic lesion.The rate of success is 66.7 per cent and the rate of mortality is 10 per cent.(2) ALT and AST go up step by step in the early stages,then go steady. TBIL increases continuously,on the contrary,albumin decreases continuously.Conclusions: (1)It can improve the achievement ratio by prolonging the period of preparation of porcine model;(2) pathology biopsy is still a gold standard of diagnosing fibrosis at present.the changes of ALT and AST have no significance for evaluating the development of hepatic cirrhosis. Albumin and bilirubin have obviously difference in before-and after-liver cirrhosis, which is useful to evaluate liver cirrhosis.(3)The feasibility of preparation of porcine model of hepatic cirrhosis by gastrostomy tube assisted with diet control is more obvious.we can hardly find any intestinal adhesion in these models,which is very meaningful to our further surgical surveys. PartⅡ:preparation of porcine model of hepatic cirrhosis by peritoneal injection of CCL4 assisted with diet controlObjective To explore the feasibility of preparation of porcine model of hepatic cirrhosis by peritoneal injection of CCL4 assisted with diet control and offer a better animal model for observing surgical therapeutic efficacy.Methods Choosing the common pig as experimental animal,the intralipid solution with 40%,CCL44 is used by peritoneal injection,0.25ml/kg and twice a week,simple corn meal was the only meal and 5%alcohol water mixture was drinked.during the course of the experimental procedure,we cut little pieces of the hepatic tissue which was prepared for HE stain and VG stain,collecting blood samples in order to test the indexes such as the function of liver and kidney,albumin,bilirubin and so on regularly.
