Purpose: study the influence of Zheng Gu Tong Bi Pills (Zheng Gu Tong Bi Wan) on the rat's osteoporosis so as to determine its curative effect and discuss its action mechanism.Method: choose the Wistar rats to form the animal models of glucocorticoid induced osteoporosis and ovary removal induced osteoporosis with the methods of eight weeks'injection of Dexamethasone and two-side ovary removal resection respectively. After modeling, continuously administrate drug by drenching for 11 or 12 weeks, then put them to death and get the thigh bone to do the tests of bone density, bone content, bone mechanics and etc; sample blood and urine, and determine the content of serum ALP, serum StrACP, urine HOP, serum OC, blood Ca, blood P, serum TNF-αand IL-6.Result: glucocorticoid induced osteoporosis model: compared with the normal reference group, the thigh bone density, bone mineral content, max. bending capacity, bending strength and break deflection of the rats in the model group all reduce obviously; the urine HOP content rises obviously; the serum StrACP increases obviously and the serum ALP reduces obviously at the same time. The dosage of Zheng Gu Tong Bi Pills dependently antagonizes the reduction of serum ALP and increase of serum StrACP caused by Dexamethasone. Compared with the model group, the group with 5.4g·kg-1 dosage of Zheng Gu Tong Bi Pills may obviously antagonizes the reduction of bone density caused by Dexamethasone, the biomechanical performance of the group with 5.4g·kg-1 dosage of Zheng Gu Tong Bi Pills obviously strengthens, and the break deflection of the groups with 2.7·kg-1 and 1.35g·kg-1 dosage of Zheng Gu Tong Bi Pills both increases obviously; and the urine HOP content of the group with 5.4 g·kg-1 dosage of Zheng Gu Tong Bi Pills obviously reduces. Ovary removal osteoporosis model: compared with the sham-operation group, the thigh bone density, bone mineral content, bending strength, max. bending capacity and break deflection of rats in ovary removal group obviously reduce; serum ALP, serum StraACP and OC rise obviously; blood Ca and P have no obvious change; the content of serum TNF—αand serum IL-6 rises obviously. Compared with the ovary removal group, the bone density and bone mineral content of each group of different dosage of Zheng Gu Tong Bi Pills increase obviously; The bone Ca, P, max. bending capacity, bending strength and break deflection of the group with 5.4g·kg-1 dosage of Zheng Gu Tong Bi Pills obviously increase. The dosage of Zheng Gu Tong Bi Pills dependently antagonizes the increase of serum StraACP and serum ALP caused by ovary removal, and the OC content of each dosage group reduces obviously; the dosage of each group with different dosage of Zheng Gu Tong Bi Pills can dependently reduces the content of TNF—αand IL-6 in rats.Conclusion: Zheng Gu Tong Bi Pills can effectively increase the bone density of the rats with the glucocorticoid induced and ovary removal induced osteoporosis, improve the biomechanical performance and strengthen the anti-fracture capacity, whose mechanism has relation with the inhibit of IL-6 and TNF—αexpression, the inhibit of bone absorption, the promotion of bone formation, and the maintenance of bone turnover balance.
