Huoxue Tablet Anti-immune Induced Hepatic Fibrosis In Rats

Objectives:To review and summarize the current understanding and treatment of traditional Chinese and Western medicine on hepatic fibrosis, to observe Huo Xue pill's influence on the immune hepatic fibrosis rat's body weight, liver and spleen weight, liver function (ALT, AST, TBA) and pathological changes of liver tissues induced by porcine serum, to observe anti-fibrosis effect of Huo Xue pill (HXP). Methods:100 Vistar female rats were randomly divided into five groups (n=20). In model group, porcine serum 0.5 ml was injected into abdominal cavity, twice per week, incessantly for ten weeks. The rest four groups, except for normal control group which was through intraperitoneal injection of normal saline, were given the same as model group and gastric perfusion with low concentration of Huo Xue pill (twenty-fold dose of adult recommended dosage), high concentration of Huo Xue pill (thirty-five fold dose of adult recommended dosage), and colchicines tablets (thirty-five fold dose of adult recommended dosage), respectively for once per day. For model and normal control groups, they were administered with distilled water 2ml by gavage. Adjustment of the amount of dose was based on weight of rats each week.At the end of 10 weeks, stop modeling.10 rats from each group were sacrificed. In order to observe conditions of body weight, weight of liver and spleen, ascites, liver function and pathological changes of liver tissue, a series of preparations should be done, like Weighing rats, fixing in supine position after anesthetization of 2% pentobarbital, taking out blood from carotid artery, opening abdominal cavity to observe ascites and to weigh liver and spleen, obtaining 1.5cm* 1cm* 0.5cm size or so liver tissue in the same position in the right lobe of liver fixed by formalin, embedded sections by paraffin,and stained by H.E.The rest rats from each group stopped modeling, and continued the above drug treatment for ten weeks. At the end of 20 weeks, they would be sacrificed, and the concrete operation was the same as the end of 10 weeks'. Not only observing conditions of body weight, weight of liver and spleen, ascites, liver function, and pathological changes of liver tissue, but also comparing indexes with the end of ten weeks'. Criteria for grading and staging of chronic hepatitis were based on the revised pathological diagnostic criteria of chronic hepatitis (seen in attached table 1) in China, which was on the foundation of Scheuer programs. The liver tissue stained with H.E., its pathological changes were observed under 60 times light microscope.Results:When the experiment was over, there was no obvious ascites in each group. There were no significant differences in body weight, weight of liver and spleen of each group in both 10 weeks and 20 weeks (P>0.05). At the end of 10 weeks, the indexes of AST, ALT, TBA of model group with liver fibrosis were all significantly higher than normal control group (P<0.01). Those indexes in low concentration of Huo Xue pill group were higher than normal group (P<0.05). In high concentration of Huo Xue pill group, there were no obvious variance compared with normal control group(P>0.05). Those indexes in high concentration of Huo Xue pill group were remarkably lower than colchicine tablet group (P<0.01). There was no obvious difference in between low concentration of Huo Xue pill group and colchicine tablet group (P>0.05).At the end of 20 weeks, the indexes of AST, ALT, TBA of model group with liver fibrosis were significantly higher than normal control group (P<0.01). There was no obvious difference in low concentration of Huo Xue pill group, high concentration of Huo Xue pill group, and normal control group (P>0.05). Compared with colchicine tablet group, those indexes in low concentration of Huo Xue pill were no great variance (P>0.05). The index of AST in high concentration of Huo Xue pill was lower than colchicine tablet group (P<0.05). The index of AST and TBA of rats in 20 weeks in low concentration of Huo Xue pill group and high concentration of Huo Xue pill group is less than 10 weeks (P<0.05). ALT was no great variance (P>0.05).At the end of 10 weeks, there were a few pathological changes, such as structural disorder of hepatic lobule, obvious expansion of portal tracts, wide fibrous septa, dense collagen, and part pseudolobular formation and so forth. Among the groups of colchicines tablet, low and high concentration of Huo Xue pill, the slight fibroplasias and few and thin fibrous septa could be found. At the end of 20 weeks, compared with 10 weeks, the liver histopathology in model group was no significant change, with pathological changes like structural disorder of hepatic lobule, expanded portal areas, part pseudolobular formation, remarkably reduced fibroplasias in high concentration of HuoXue pill group, and inflammation in part of portal tracts, etc.Conclusion:Huo Xue pill can restrain the rat's immune hepatic fibrosis induced by porcine serum, efficacy and dosage have correlationship with treatment time and potency。...
