Purpose:To discuss intervention effect and mechanism of Buyuancongnao decoction on Alzheimer's disease(AD) by observing the decoction's effect on Alzheimer's disease model rats about superoxide dismutase (SOD),malondialdehyde (MDA),nitric oxide synthase (NOS) and so on。Material and method:There were 75 healthy SD male rats,weight (280±20) g,provided by animal experiment center of LNUTCM。The feeding room temperature was 20℃,the rats could get water and feed freedomly。After 2 weeks of adaptive feeding,the rats whose reaction was too slow or sensitive were washed out by the Morris water maze。The remaining rats were randomly divided into normal group (15 rats),sham operation group (14 rats) and model group (41 rats)。The AD rats model was made by injecting the mixture ofβ-amyloid protein (Aβ1-42) and ibotenic acid (IBO) in the rats'hippocampal CA1 area in model group。All the rats in model group were tested with Morris water maze after modeling, and a total of 37 rat model was successful。Then the 37 model rats were randomly divided into 3 groups:model control group,Chinese medicine group and western medicine group,12,12,13 each group respectively。The rats in Chinese medicine group and western medicine group were treated with Buyuancongnao decoction and Piracetam Tablets respectively。The learning and memory of rats was judged by Morris water maze,the pathological changes of the hippocampus were observed under light microscope,and the changes of SOD,MDA,NOS were detected。The datas were statistically analyzed by spss15.0 package。Results:1.Compared with the normal group,swim time and the percentage in total time of model group rats in the former platform quadrant reduced (P<0.05)。It showed that the learning and memory of these rats were significantly decreased。This also meant that the model was successful。After treatment with drugs swim time and the percentage in total time of rats in Chinese medicine group and western medicine group in the former platform quadrant increased (P<0.05)。It showed that the learning and memory ability of these rats got significantly improved。The difference of effect between two groups wasn't significant(P> 0.05)。2.Compared with the normal group,the rats'SOD activity reduced,serum MDA content increased,NOS immunoreactive neurons count was increased in model group (P<0.05)。Compared with the model group,after intragastric administration,the rats'SOD activity raised,serum MDA content was decreased,NOS immunoreactive neurons count decreased in treatment group (P<0.05)。It showed that the learning and memory ability of these rats in treatment group got significantly improved。There was no significant difference between Chinese medicine and western medicine group (P> 0.05)。3.Compared with the normal group,the number of neurons model group significantly reduced,but the number of positive neurons significantly increased。Such cell apoptosis appearance as part of the neuronal nucleus pyknotic,nuclear membrane thickening and aberration,even neuronal degeneration or necrosis and lipofuscin increased could be observed。The number of positive neurons was decreased in Chinese medicine and western medicine group。And the distribution of nerve fibers in the two groups was more regular than the model group。There was no significant difference between Chinese medicine and western medicine group。Conclusion:1.The AD rats model,made by injecting the mixture of Aβ1-42 and IBO in the rats'hippocampal CA1 area,was a more reliable animal model。It could successfully simulate the behavior and pathological features of Alzheimer's disease。2.Buyuancongnao decoction could improve the learning and memory ability of AD rats。3.Buyuancongnao decoction could improve the learning and memory ability of AD rats by reducing serum MDA content and NOS positive neurons count,increasing SOD activity。...
