Yagong Leaf Chemical Composition And Quality Standards Study

Purpose:To study the chemical composition of the leaves of Smallanthus sonchifolius (Yacon) and examine the contents of chlorogenic acid and total phenolic acid in the leaves of Smallanthus sonchifolius (Yacon) cultivated from different places.Material and method:Various kinds of chromatography methods, such as silica gel, ODS, Sephadex-LH20 column and preparative HPLC, were used in the isolation procedures, while the structures of isolated compounds were determined on the basis of NMR and MS spectral analysis. The content of total phenolic acid in the leaves of Smallanthus sonchifolius of different places was determined by colorimetry. HPLC method was used to determine of the content of chlorogenic acid in the leaves of Smallanthus sonchifolius from different places.Results:1. Eight compounds were isolated from its water and ethanolic extract:Smaditerpenic acid B (1), Smaditerpenic acid D (2), Benzyl alcohol 7-O-α-D-arabinopyranose (1〞â†'2ËŠ) -β-D-glucopyranoside(3) , 3-Hexylene-O-α-D-arabinopyranose(1〞â†'2ËŠ )-β-D glucopyranoside(4),Grandiflorolic acid (5),13(R)-hydroxy-octadeca-(9Z, 11E, 15Z)-trien-oic acid (6) , 13(R)-hydroxy-octadeca-(9E,11E,15Z)-trien-oic acid(7) , ent-18-hydroxy-kauran-16-en-19-oic acid (8)。2. The content of chlorogenic acid in the leaves of Smallanthus sonchifolius from Anshan (China) was 0.3621mg/g. And the content of chlorogenic acid in the leaves of Smallanthus sonchifolius from Yunnan was 0.1266mg/g, and from Dalian was 0.1188mg/g.3. The content of total phenolic acid in the leaves of Smallanthus sonchifolius changed obviously, the content of total phenolic acid in the leaves of Smallanthus sonchifolius from Anshan was 5.82%, the next was from Benxi was 5.26%, and from Yunnan was 4.92%, and from Dalian was 4.24%.Conclusions:1. The content of Chlorogenic acid in the leaves of Smallanthus sonchifolius from Anshan (China) was higher than that from Yunnan and Dalian.2. The content of total phenolic acid in the leaves of Smallanthus sonchifolius from Anshan were the highest, the next was from Benxi, from Yunnan and Dalian were lower.
