Methods:64SD rats were randomlydivided into four big groups: The saline control group (NNa group)、The pretreatmentof aerobic exercise before acute aging group (SD group)、The acute aging group (NDgroup)、The acute aging process aerobic exercise intervention group (NDs group).After modeling of aging, each group was randomly divided into two groups: A Morriswater maze test done7days for group M, Another group made seven days of naturalfeeding, as N group(For example: NNa group was divided into groups NNaM andNNaN group), A total of eight group. Expression detected by immunofluorescence,Western-blotting, real-time PCR experiments in rats BDNF gene in the hippocampus.Result:1. The appearance of morphological: Acutely aging rats compared with thecontrol group of rats, Appeared in the spirit of the poor、Unresponsive、Sleepiness、Fur lackluster, curly withered and yellow, Severe hair loss and other symptoms ofaging significantly.2. Detection of free radicals: NNaN group of gsh-px and SODactivity significantly higher than that of NDN group(P<0.01),While NNaN MDAcontent significantly lower than NDN group(P<0.01);NDN group and SDN MDAcontent and SOD activity of gsh-px and there was no significant difference(P>0.05);NDN group of SOD activity was very significantly lower than that of NDsN(P<0.01),Gsh-px activity significantly lower than that of NDsN group (P <0.05), MDA contentis very significant higher than that of NDsN group (P <0.01); NNaN and NDsNgroup of MDA content and gsh-px and SOD activity were no significant difference (P>0.05). Comparing four group M, Each change trend and the N group of basicallythe same. The N group compared with the corresponding group M MDA content andSOD activity of gsh-px and no significant difference (P>0.05).3. Morris water mazeof navigation experimental results: NDM, SDM, NNaM, NDsM four groups of ratsare trained in2days after the initial formation of the spatial learning and memory, Stable spatial learning and memory of rats NDM group is formed after4days training,SDM group of memory formation is unstable, volatile, Stable spatial learning andmemory of rats NNaM group is formed after training in3days, Stable spatial learningand memory of rats NDsM group is formed after the3days training. Morris watermaze of space exploration results: Compared with NNaM group, NDM group ratsfirst entry quadrant platform through the number of times very significantly lower (P<0.01); NDM group compared with SDM group, there was no significant difference(P>0.05); NDM group compared with the NDsM group was significantly lower (P <0.05).4、Immunofluorescence test:BDNF was massive distributed in the cytoplasmof neurons.NNa and NDs group, positive cells number, strong positive expression.ND and SD group, the decrease in the number of nerve cells, less number of positivecells, weakly positive expression.5、The hippocampus BDNF mRNA and proteindetection:NDN group compared with NNaN, the hippocampus BDNF mRNA andprotein significantly lowered(P<0.01);SDN group compared with NDN group, thehippocampus BDNF mRNA and protein there was no significant difference(P>0.05);NDsN and NDN group compared to the hippocampus BDNF mRNAandprotein expression significantly raised(P<0.01);NDsN group compared with NNaNBDNFmRNA there was no significant difference compared with the amount ofprotein expression(P>0.05)。Comparison between four M group, the change trend ofBDNF mRNA and protein are the same as the N group. Four M and thecorresponding N each group comparison, mRNA and protein expression had nosignificant difference(P>0.05)。 Conclusions:1. After Injection of D-galactose onrats to aging, In the aging rat SOD, gsh-px activity significantly lower than that ofnormal saline control group, MDA content significantly higher than that of normalsaline control group, And appear obvious symptoms of aging. That the experimentalD-galactose building successful brain aging rats.2. Exercise pretreatment beforeaging rats is no significant meaning on the influence of spatial learning and memoryability; Aerobic exercise intervention in the process of aging rats, Can effectivelyslow brain aging process, Aerobic exercise on the organic positive impact may be asthe movement to stop slowly fade.3. During the senescence of aerobic exerciseintervention can maintain the normal hippocampus BDNF mRNA and protein expression, This may be aerobic exercise reduce rats learning and memory ability isone of the reasons for decline.
