Research purposes:(1)To explore the role of BDNF/CREB pathway in motor impact cognition;(2)To explore the impact of treadmill exercise on hippocampal neurogenesis and learning and memory abilities,and whether there is a gradient effect between different intensities;Methods: Forty one-month-old male SD rats were randomly divided into control group(Con),small intensity group(LE),medium intensity group(ME)and high intensity group(HE).The control group(Con)received standard feeding,and the exercise group received treadmill intervention according to the corresponding intensity for 2 months.After the intervention,the spatial learning and memory abilities were evaluated by Morris water maze(MWM);The hippocampus tissues of rats were dissected,and DCX,Neu N and Ki67 positive cells were detected and observed by immunofluorescence and immunohistochemical methods to explore the neurogenesis;Western blot and PCR were used to detect the protein expression and gene level of BDNF/CREB,the potential mechanism of hippocampal neurogenesis.Results:(1)Morris water maze: Compared with the Con,the escape latency in the exercise group was significantly reduced(P<.05),and there was no significant difference between different intensities(P>.05);Compared with HE,LE and ME had shorter residence time in platform location(P<.05);Compared with Con group,the residence time in HE was significantly increased(P<.01);Compared to Con and LE,the target quadrant residence time in ME was significantly increased(P<.05);(2)Immunofluorescence and immunohistochemistry: Compared with Con,Neu N+and Ki67+ neurons in exercise group was significantly increased(P<.05);There was no significant difference between different intensities(P>.05);(3)Western Blot: Compared with Con,the BDNF and CREB protein level in LE and ME were significantly increased(P<.01);Compared with LE and ME,CREB level in HE group was significantly decreased(P<.05).(4)PCR quantification: Compared with the Con,the BDNF m RNA transcription level in exercise group was significantly increased(P<.05);Compared with Con,the level of CREB m RNA gene transcription in ME was significantly increased(P<.05).Conclusion:(1)Treadmill exercise can significantly improve the spatial learning and memory,with a certain gradient effect between different intensities;(2)Moderate and high intensity exercise can significantly increase the positive expression of Neu N and Ki67 in the hippocampus and promote hippocampal neurogenesis;(3)Moderate intensity exercise can induce morphological changes in the hippocampus by regulating BDNF/CREB pathway,and improve cognitive ability.
