It is discovered that the free radical in body was increasing during the long-term high intensity and endurance-training, which breaks the balance between the production and elimination of the free radical. Furthermore, the concentration of the free radical exceeds the normal level, which induces broad damnification in cells, tissues and some pathology turbulence. Too much free radical may attract polyunsaturated fatty acid on the biology membrane and form lipid peroxidation, which induce a series of pathology and physical change in the structure and function of biology membrane. It decreases work capability and makes people tired. In addition, as is shown in the investigation that over accumulation of ROS may induce apoptosis. Apoptosis may be caused by the midway production of ROS, Many physical and chemical factors, which induce apoptosis, can arouse producing the free radical too. The unbalance of the production and elimination of the free radical can cause apoptosis. ebselen is a kind of small molecule antioxidant, and is accepted as the good simulacrum of GSH-Px, whose toxicity is low, and easy to take part in any kind of oxidation-deoxidization reaction. It can eliminate the over-peroxid in the body, interdict the chain reaction that can produce the free radical, and maintain body's normal physical function. Many scholar's study shows that ebselen has obvious effect on the aspects of biology anti-oxidation and anti-inflammation. It also can control apoptosis and relieve the damnification of heart, liver and brain nerve caused by stimulation. In order to provide experiment basis for the use of ebselen in the sports medicine field, this experiment observes the free radical metabolism and cell morphologic effect of some tissues of high-intensity and endurance-trained rats taken ebselen.This experiment uses 50 SD healthy male rats, whose weight are from 180g to 220g. They are divided into three groups at random: 8 rats for quiet group; 16 rats for exercise contrast group; 16 rats for exercise drug-taken group. At the last time for training, 8 rats are chosen from exercise group as exercise exhaust group and chosen another 8 from exercise drug-taken group as drug-taken exhaust group. On the last dayof the 8th week of the training period, the exhausted time of both exercise exhaust group and drug-taken exhaust group should be recorded. Then, all of the objects are weighted, hocused by aether and taken the materials from some tissues. This experiment tests MDA, SOD, GSH-Px, GST, CAT and LDH, which can reflect the defense system of anti-free-radical and damnification condition in the economy; NOS, NO etc that reflect the index of N-free-radical containing in the body, and bcl-2, bax's protein expression; observes and tests the apoptosis of some tissues by TUNEL method; and some tissues through the transmission electron microscope.Result: a) Ebselen can extend the exhaust time of rat of treadmill training with high-intensity and endurance-training, which has anti-fatigue function. b)Ebselen can postpone exercise fatigue induced by over-produced free radical because it can reduce the content of MDA in the heart, liver, muscle, kidney, brain and blood of high-intensity and endurance-trained rats distinctly, improve the enzymatic activity of SOD, GSH-Px, GST, CAT, etc, decrease the activity of LDH and improve the activity of T-AOC in the economy, c) Ebselen can decline the activity of NOS and iNOS in those tissues, and the production of NO of rats during the endurance-training, prevent the production of N-free-radical during training, restrain the apoptosis, and postpone exercise fatigue, d) Ebselen can increase the expression of bcl-2 protein and decrease bax protein, which make the ratio of bax/bcl-2 decrease. It goes against the apoptosis during exercise, then postpone fatigue; the number of apoptosis cell of drug taken group according to TUNEL. e) By observing through transmission electron microscope, ebselen protects the cardiac muscle, skeleton muscle and hepatic structure of high-intensity and endurance- trained rats...
