The purpose of the experiment:To explore the relationship between high-fat diet induced testicular germ cell apoptosis and inflammatory factor TNF-alpha, IL-6 on male SD rats, and to reveal intervention effects of aerobic exercise on testicular germ cell apoptosis and inflammatory reaction in high-fat diet rats.The Method of the experiment:Totally 48 healthy male Sprague-Dawley (SD) rats were randomly divided into 4 groups: normal control group (C), high fat diet group (HF), general exercise group (CE), high fat diet and exercise group (HE),12 rats in each group; C group and CE group were fed with common forage, HF group and HE group were given high fat, high heat. After the end of the experiment, the number of rats in each group:C Group 8, HF group 11, CE group 10, HE group 11. Kinematic scheme for 10 weeks 60 min without weight loading swimming movement, six days a week. Test after test in rats serum triglycerides (TG), total cholesterol (TC). high density lipoprotein (HDL), low density lipoprotein (LDL); Measurement of rat body weight, body length, fat weight, calculation of Lee’s index and body fat ratio; Enzyme linked immunosorbent assay (ELISA) determination of tumor necrosis factor alpha (TNF-alpha), interleukin 6 (IL-6); RT-PCR to detect epididymal fat TNF-alpha, IL-6 mRNA and TNFR, IL-6R mRNA in testis; the morphological structure of the testis was observed with HE staining; In situ nick end labeling (TUNEL) to detect apoptosis of testis spermatogenic cells in rats。The result of the experiment:1.After 10 weeks, CE group compared with C group, body weight was significantly decreased. Lee’s index, TG significantly reduced, HDL was significantly increased, epididymal fat, body fat ratio, TC, LDL had no significant difference. HF group compared with C group, the body weight, epididymal fat, body fat, TC, LDL was significantly increased, no significant differences were found in TG, HDL, Lee’s index.2.CE group compared with C group, in rat serum TNF-alpha, IL-6 levels were significantly lower. HF group compared with C group, TNF-alpha levels in serum of rats increased significantly, the level of IL-6 increased, but no significant difference. HE group compared with HF group, TNF-alpha, IL-6 levels significantly reduced. Through the single factor variance analysis, high fat diet can induce the levels of TNF-alpha and IL-6 increased in serum of rats, and aerobic exercise can decrease the serum level of TNF-alpha and IL-6.3.Morphological observation of rat testis by HE staining under light microscope, found in the C group within the seminiferous tubules spermatogenic cells of normal development at all levels, arranged in neat rows, bureaucratic without obvious stenosis and dilated, the lumen can be seen a large number of mature sperm. CE group in the seminiferous tubules of the testis gap widens, mature sperm uniform distribution. Testis tissue in HF group of seminiferous tubules wall thinning, the number of spermatogenic cells was significantly reduced and arranged in disorder, the number of mature spermatozoa significantly reduced, spermatogenic cells appeared off phenomenon, between individual rats spermatogenic cells and basement membrane appeared out of. HE group in seminiferous tubule spermatogenic cell the basic development normal, even visible cell shedding phenomenon, duality can be born seminiferous tubule interstitial tissue is loose.4.CE group compared with C group, the rat epididymal TNF-alpha, IL-6 mRNA levels increased, but no significant difference; HE group compared with HF group, epididymal TNF-alpha, IL-6 mRNA levels decreased significantly. Through the single factor variance analysis, high fat diet can improve the epididymal TNF-alpha, IL-6 mRNA levels of rats, epididymal fat induced inflammatory response, and aerobic exercise can be reduced TNF-alpha, IL-6 mRNA levels, the effect of reducing inflammation.5.CE group compared with C group, the testis tissue in rats TNFR, IL-6R mRNA levels was increased, but no significant difference; HE group compared with HF group, TNFR, IL-6R mRNA levels in testis tissue was significantly decreased. Through the single factor variance analysis, high fat diet to make TNFR, IL-6R mRNA level in rat testis significantly increased, causing testicular tissue inflammatory response, and aerobic exercise can be reduced TNFR, IL-6R levels of mRNA, effect of reducing inflammation.6.Cell apoptosis situation in rat testicular tissue:compared with C group, germ cell apoptosis rate of testicular tissue increased in CE group, but no significant difference, germ cell apoptosis significantly increased rate of testicular tissue in group HF and group HE. HE group compared with HF group, apoptosis of germ cells significantly decreased the rate of testis. Through the single factor variance analysis, high fat diet may cause inflammation of testicular tissue, resulting in testicular germ cell apoptosis in tissue increased, while aerobic exercise can alleviate this situation.Conclusion:1.High fat diet can cause the disorder of fat metabolism in male SD rats, the protein expression level of TNF-alpha, IL-6 in serum and mRNA in the epididymal fat with Lee’s index and four blood lipids have correlation,and aerobic exercise can improve the situation.2.High fat diet can make the structure of testes in SD male rats changes, and make the sperm apoptosis increased significantly, through aerobic exercise can make the situation improved.3.High fat diet can cause the increase of rat serum TNF-alpha, IL-6 protein expression and epididymal fat mRNA and testicular tissue TNF-alpha and IL-6 mRNA levels, the mean and the rate of apoptosis is closely related to, and aerobic exercise can inhibit effect, which may also is the molecular mechanism of aerobic exercise intervention of obesity inflammatory rat germ cell apoptosis.
