Objective: Through the establishment of aging SD rats(N=60) animal model, to intervene the animal model with chronic exercise training, in order to explore the different stages in the course of aging the physiological changes of skeletal muscle cells, and then reflecting the influence of exercise on body skeletal muscle cell oxidation-anti oxidation dynamic balance and the expression characteristic and the expression level of cell apoptosis related proteins, thereby to explore sports fitness value and the influence of exercise on skeletal muscle cell biology mechanism. Methods: This experiment adopts the three ages of SD male rats as experimental research objects, randomly divided into control group and experimental group, according to the age-related changes are divided into: young group(N=10), middle age group(N=10) and the elderly group(N=10), treadmill training: the slope of 0 degrees, speed 15 m / minute, 60 minutes, training 8 weeks. After the training, rats were anesthetized and extract femoral head four muscle tissue, in accordance with the needs of the experiment test, make the corresponding sample, preservation for testing. Detection of oxygen free radical metabolism using kit, SOD by xanthine oxidase method; MDA by thiobarbituric acid method, at the same time with the immunohistochemical method for Bcl-2 and Bax gene expression in skeletal muscle tissue The conclusion of the study:1. The expression of apoptosis related protein was influenced by age factors. Under the influence of aging factors, each age group of rat skeletal muscle all exist metabolic changes of oxygen free radical, and this kind of change gradually increased with the age increasing; Expression of apoptosis related proteins in skeletal muscle tissue levels did not show a linear relationship, but in the elderly group changed obviously(P<0.05).2. The occurrence of regular exercise can significantly induce apoptosis phenomenon, the extent of exercise intervention on apoptosis induced in elderly patients bcl-2 most significant(P <0.05), the young and middle-age groups in obvious influence; Skeletal muscle tissue after exercise each group SOD activity was significantly increased, while the MDA content in skeletal muscle cells in the older group showed a rising trend(P <0.05).3 Moderate exercise has beneficial effects on the body which has been widely known. The experiment results show that, 8 weeks of treadmill exercise can adjust the body to produce and elimination ability and the oxygen free radicals, it also improved the antioxidant capacity of skeletal muscle cells, reducing the possibility of free radical damage cells. And in elderly apoptotic Bcl-2 gene expression in the ability to influence more significant, this delayed the physiological function of skeletal muscle tissue of progressive decline.
