Objective:The aim of this study is to observe cell morphology of rat soleus and analyse the change of IL-6 and TNF-a in plasma and soleus, to explore the influence of acupuncture intervention on inflammatory response of skeletal muscle after eccentric exercise and the function of IL-6.Methods:8 weeks age male Spraque-Dawley (SD) rats total 114, were randomly divided into four groups:control group(C), eccentric exercise group(E), acupuncture group (A,) and sports acupuncture group (EA), control group and sports acupuncture group rats were submitted to eccentric exercise at a speed about 16 m/min on a treadmill down a 16°decline for ninety minutes, eccentric exercise group and sports acupuncture group for oblique thorn oftriceps surae, acupuncture to rats soleus for 2 minutes, then gaining the issue according to the Oh,2h,24h,48h,72h,120h phase. Observed cell morphology of rat soleus with HE, examined IL-6, TNF-a protein in soleus with Western Blot; The plasma IL-6 and TNF-a levels were determined by Elisa.Results:(1) HE results,acupuncture group and control group showed no significant difference, exercise group with varying degrees of inflammatory cell infiltration, sports acupuncture making inflammatory cell infiltration of the peaks ahead. (2) In the soleus, acupuncture group IL-6 and TNF-a showed no significant difference compared with the control group; 2h peak of IL-6 higher than at the same time phase in sports acupuncture Group (P<0.05). TNF-α,eccentric exercise group reached its peak in 72h,and sports acupuncture group reached its peak in 48h. (3) IL-6 and TNF-a in plasma, acupuncture group compared to the control group showing no significant difference. IL-6 eccentric exercise group reachued the highest sports in 2h, and higher than sports acupuncture group at the same phase, but no significant difference (p>0.05).TNF-a in eccentric exercise groups immediately reached to the highest peak and sports acupuncture group reached its peak in 2h.Conclusions:1. In this model, the eccentric group has no important muscle inflammation for experimental results, including eccentric exercise group causing muscle inflammation, and compared with eccentric exercise group,the sports acupuncture group accelerate the process of inflammation reaction.2. Acupuncture by inhibition of myogenic IL-6 in the 2h, reducing inhibition of TNF-α, this is a part of the mechanism for impact of acupuncture on inflammatory response of skeletal muscle.
