| Objective: Explore the SD rats in the long-term intervention in different exercise stress on learning and memory capacity and prefrontal OX1 R gene expression levels.Methods:The 30 male SD rats were divided into three groups, Control Group(group C) 10 rats;Medium load Group(group M)10 rats; Excess load Group(group C) 10 rats.Three groups of rats were beginning to behavioral test one day to rule whether its inherent differences in learning ability.Eight weeks in the control group fed natural.Medium load Group adaptive swimming for three days, for eight weeks of swimming training,swimming training before the first 10 min / day in rats after 1/6 days / week,formal training rats 1st water swim 10 minutes, after which the daily cumulative 5min, a weekend trip to the first 30 min,tour to the second weekend is just 60 min, beginning from the first three weeks of daily cumulative 10 min, to the third weekend just swim 120 min, the load continues to 8 week tour.O groups of eight weeks of swimming training, the first three weeks with M group,from the beginning of the first four weeks load 1% of their body weight in rats, an increase of the first five weeks to 2% of their body weight load, the first 6 weeks to 3%, the first seven weeks of negative 4 percent of their body weight, the first eight weeks of their body weight minus 5 %, 120 min / day.Three groups of rats after 8 weeks Morris water maze behavioral training, to adapt water maze environment one day(free swimming 2 minutes), the water maze training seven days and behavioral test one day, were recorded for the group C, M and O groups groups.After decapitation drawn, use Real-time PCR, Western-blotting OX1 R measured expression of the three groups of rats frontal lobe. Results: 1ã€The general observations showed M group after exercise will accelerate the respiratory rate performance, mental everything is normal, but O group movement emerged after the excessive load limp, dull eyes, respiratory rate rapidly speeding up.2ã€Prefrontal Real-time PCR results showed that:OX1R expression of O and M group were significantly higher than group C(P <0.05), whereas the expression of M O group than the group;Prefrontal Western-blotting results showed that:M group was significantly lower than that of group C(P < 0.01) and O group, OX1 R expression quantity O group is not particularly significant difference compared with group C.3ã€Immunofluorescence results show that the M group, O group, the number of positive cells express less and less positive, M group and O group difference was not significant compared with group C.4 〠Morris water maze experiment positioning voyage, M group, the average escape latency time was significantly lower than that of group C(P <0.01);O group with no significant difference between group C.Space exploration experiments, the number of cross platform between O group and C group no significant difference(P> 0.05).About four days after the first two groups of rats were entering the plateau, and the average escape latency time between the two is basically the same.After which the number of the second quadrant into the water, O group, the average cross-platform significantly lower than in group C(P <0.05) and M groups.Conclusion:1ã€Moderate exercise load can significantly improve learning and memory in rats, and excessive exercise stress on learning and memory in rats and no significant impact.2ã€OX1R different expression of the frontal lobe by the exercise load size and have different levels of expression, medium load has reduced the amount of OX1 R expression, thus reducing the amount of expression in the frontal OX1 R site may be an improvement on learning and memory abilities important factor, so moderate Exercise can learn expression in rat brain and memory-related proteins have improvement. The influence of excessive exercise load on the expression of OX1R’s not obvious, so the learning and memory in rats and there was no significant improvement. |
