Objective:Through the different load intensity swimming exercise intervention in rats,we observed the movement influenced on rat spatial learning and memory ability and detected the rat hippocampal proteins expression which associated with learning and memory(NCAMã€OXAã€OX1R).Methods: 30 2-month-old male Sprague-Dawley rats were selected,fed for adaption for 3 days.Then randomly divided into three groups:Control Group(C,n=10),Medium load Group(M,n=10),Over load Group(O,n=10).C group feeding 8 weeks naturally, M group do swimming exercise intervention for 8 weeks, O group do over load swimming exercise intervention for 8 weeks.Morris water maze system evaluated the effects of different periods of different exercise on spatial learning and memory abilities.Put rats to death according to the building time and take brain and blood.Detected blood serum testosterone content by ELISA technique;Detected the m RNA expression level by Real time PCR;Detected the protein expression level by Western blot. Results:(1)Body weight monitoring results:since the end of third week average weight of M and O group were lower than that of C group(P<0.01),since the end of fourth week the weight of O group was lower than M group(P<0.05);(2)Blood testosterone test results:there was no significantly difference between M and C groups(P>0.05),the blood testosterone of O group rats was significantly lower than M and C groups(P<0.01);(3)Results of Morris water maze:during the navigation experiment,M group was easier to find the platform than C group,and the fluctuation range of time to find platform is small(P<0.01).During the space exploration,the M group through the platform number was significantly more than C group(P<0.05),O group through the platform number was significantly more than C group and M group(P<0.01,P<0.05);(4)Results of hippocampal related protein m RNA test:the expression of NCAM m RNA and OXA m RNA of M group were significantly higher than C group(P<0.05),the expression of OX1 R m RNA was significantly lower than C group(P<0.05);(5)Results of hippocampal related protein test:the expression of NCAM of M group were significantly higher than C group(P<0.05),the expression of NCAM of O group were significantly lower than C group(P<0.05),the expression of OXA and OX1 R of M group were significantly lower than C group(P<0.05);(6)Results of every index’s correlation test:the through platform number,the expression of NCAM m RNA,NCAM,OXA m RNA of hippocampal had significantly negative correlation with average latent(P<0.05,P<0.01),the expression of OX1 R of hippocampal had significantly positive correlation with average latent(P<0.01);the expression of NCAM and OXA m RNA of hippocampal had significantly positive correlation with the through platform number(P<0.05,P<0.01),the expression of OX1 R of hippocampal had significantly negative correlation with the through platform number(P<0.01);the expression of NCAM m RNA had significantly positive correlation with the expression of OX1 R of hippocampal(P<0.01),the expression of NCAM had significantly negative correlation with the expression of OX1 R and OXA of hippocampal(P<0.01).Conclusion:1. Medium load exercise could improve the performance of rats in Morris water maze experiment,and improve the rats spatial learning and memory ability,medium load exercise could significantly improve the expression of OXA,OX1 R m RNA and OX1 R,significantly reduced the expression of OXA,OX1 R m RNA,OX1 R.Perhaps the OX1 R signal could influence the expression of NCAM to regulate and influence learning and memory ability. 2. Over load swimming exercise could damage the rats spatial learning and memory ability.After over load swimming exercise,the expression of NCAM was normal perhaps related to its function of nerve repair,the expression of OXA and OX1 R was normal perhaps related to its special bi-stable adjustment,its mechanism need to be further discussed. |
