Objective: Selecting the person that is simple obesity, to construct the genomic expression profile of adipose tissue using the serial analysis of gene expression method, describe the transcriptome and distribution.Research Methods and Procedures: Subcutaneous adipose tissue was obtained from one male adult patient with simple obesity. The mRNA were extracted and kinds of transcripts were analyzed using SAGE, and then the expression counts were defined. The serial analysis of gene expression strategy is based on isolation of short sequences (tags),which usually correspond to unique transcripts; and on their concatenation into long DNA molecules, which are then cloned and sequenced using SAGE software. The sorts of transcripts were identified. The expression counts were calculated by the frequency of tags. Classification of the genes related to metabolism was based on the updated information of CGAP Gene Ontology Browser web site (http://www.cgap.nci.nih.gov/Genes).Results and Conclusion: We isolated 67562 tags in SAGE library. Among them 1893 tags were related to metabolism, which representing 140 different genes. And the function of 38 genes had been determined. The numbers of genes related to energy metabolism , fatty metabolism, carbohydrate metabolism, amino acid metabolism and nucleotide were 2,6,1,13 and 16, respectively. The rates occupying total genes were 5.26%, 15.8%, 2.63%, 34.2% and 42.1%, respectively.The most expressed gene is SCAP2, and the count is 0.48% of the total population. Nevertheless eleven novel transcripts were found, but were at a lower level.
