Screening of bioactive compounds in traditional Chinese medicines (TCMs) is an extremely challenge, and also it's the basic and crucial problem of the modernization of TCMs. A novel way had been provided to resolve the problem potentially with the proposing of biolofical fingerprinting analysis.The information which was responding the interaction of the components from TCMs with the biological system drawing directly from the biological fingerprinting could guide the screening of the bioactive compounds of TCMs. Two active components, which were hydroxysafflor yellow A and anhydrosafflor yellow B, of Flos Carthami had been screened with the applying of biological fingerprinting analysis in our lab. In the paper, Two bioactive components above-mentioned has been prepared and the studies on the interaction of them with human serum albumin (HSA) has been carried out.Firstly, column chromatographical method of the preparing of hydroxysafflor yellow A and anhydrosafflor yellow B has been optimized. And both of them were prepared.Secondly, the interation of hydroxysafflor yellow A and anhydrosafflor yellow B with HSA has been studied by the method of centrifugal ultrafiltration coupled with HPLC, and their binding constants with HSA has been calculated.Finally, the influence of the interaction of hydroxysafflor yellow A and anhydrosafflor yellow B with HSA by iburpofen or warfarin has been studied with the method of centrifugal ultrafiltration coupled with HPLC as well. And the binding site of Hydroxysafflor yellow A and Anhydrosafflor yellow B on HSA has also been determined.
