An aqueous enzymatic extraction technology was adopted to extract oil from Oil-tea camellia seed. In order to provide a new way for Comprehensive utilization and deep processing of Camellia oleifera Abel., the best enzymes and enzyme complex formulas on oil yield were investigated by multi-level optimization procedure, the optimum enzymatic hydrolysis conditions were gained based on response surface methodology, the mechanism of aqueous enzymatic extraction of oil-tea camellia seed oil was investigated by using TEM and SEM, the suitable demulsification methods which was the key step of aqueous enzymatic extraction of Oil-tea camellia seed oil were studied too.The influences of different enzymes and enzyme complex formulas on oil yield were investigated. The result showed that Alcalase 2.4L was the best single enzyme with the highest oil yield of 90.48%. The best enzyme complex formula was composed by pectinase, cellulase and Alcalase 2.4L , which was gained by multi-level optimization procedure. The oil yield efficiency could reach to 91.40% when this enzyme complex formula was used in sequence. By comprehensive consideration, we choose Alcalase 2.4L as the best hydrolase.Alcalase 2.4L was chosen as the hydrolytic enzyme, The influences of different enzymatic hydrolysis conditions on the oil yield were investigated. Based on response surface methodology, the optimum conditions were found as follows: dosage of the enzyme 2%, temperature 61.5℃, hydrolysis time 4h, pH value 9.4, ratio of material to water 1:7, grind degree 80 mesh. The verification test showed that the oil yield can reach 91.24% at the optimum conditions.Using TEM and SEM to observe the intracellular and extracellular changes of Oil-tea camellia seed cell during aqueous enzymatic extraction of oil from it. The result showed that the lipid granules were discrete distributed in Oil-tea camellia seed cell, and confirmed that Alcalase 2.4L could enzymolysis cell wall effectively and release cell contents including lipid granules from the morphological view. The degradation of cell wall was a dynamic process. For demulsifing the emulsion forming in the aqueous enzymatic extraction of oil from Oil-tea camellia seed. The composition of emulsion and suitable demulsification methods was studied. The result showed that using ethanol with concentration of 30% could demulsify the emulsion effectively. The verification test showed that the oil yield can reach 88% at the centrifugal condition of 12000rpm and 20min.
