This study was used oil-camellia seed as raw material to explore the influence of different mechanical treatments and different enzymes on oil-camellia seed oil processing by aqueous enzymatic method. Both orthogonal experiment and response surface design method were applied to optimize the extraction conditions of oil-tea camellia oil by aqueous enzymatic method. The main results are as follows:1.This paper studied the influence of different mechanical treatments on oil-tea camellia oil processing by aqueous enzymatic method, and observed the surface structure of cotyledon cells of oil-tea camellia seed crushed by different mechanical equipments by means of scanning electron microscopy. The results show that the raw material particle size of oil-camellia seed were mainly in 30~150μm by pulverizing mill, but most part of cells fail to destroy to effect the oil getting out. The diameter of cells are 15~40μm after secondary mill grinding, which can effectively destroy cells structure, increase the effective contact area of enzymes. It was confirmed that the oil slurry after second grinding had higher oil extraction rate.2.Different types of enzymes were screened from oil-camellia seed oil processing by aqueous enzymatic method, the results showed that oil extraction rate of high temperature amylase is the highest, followed by temperature amylase, acid protease and pectinase. With high-temperature amylase enzyme as the single enzyme, the oil-extracting yield and residual oil rate as evaluation indicators, single-factor tests and orthogonal test were carried out to investigate the oil-extracting yield at different levels on of different enzyme dose, extraction temperature, extraction time, extraction pH and feed/water ratio. The results showed that the optimum extraction parameters of enzymatic hydrolysis process were as follows:enzyme dose is 0.10%, extraction temperature 85℃, extraction time 3 h, extraction pH is 6, the ratio of material to water is 1:4. Under this condition the oil-extracting yield is higher than 92.23 %.3.Based on the oil extraction rate of camellia oil by a single enzyme species, high temperature amylase, acid protease and pectinase enzymes were selected as compoud-enzyme. The results showed that the highest extraction rate was obtained when high temperature amylase and acid protease as a combination, and adding high temperature amylase enzyme first. Single-factor tests and Box-Behnken design tests were carried out to investigate the oil-extracting yield at different levels of the amount of addition enzyme, the ratio of water, extraction time. The results showed that the optimum extraction parameters of enzymatic hydrolysis process was as follows:compound enzyme dose 0.13%, water ratio of water 1:3.9, reaction time 4h. Under this condition, the oil extraction yield was 92.45%.
