| Radix Slaviae Miltiorrhizae is dried roots and rhizomes of Salviae miltiorrhiza Bunge. Sal A (salvianol acid A, SalA) is a phenolic compound extracted from Salvia a water-soluble components. Sal (Salvianolic acid, SA) is the water soluble components extracted from Salvia which has both the structure of coffee phthalic acid and the skeleton of new lignan. It is the main active ingredients as the role of blood circulation.The active ingredients includ Salvia, protocatechuic aldehyde, rosmarinic acid, oxalic purple acid, Sal A and Sal B. However, Sal B has the highest concentrations in Radix Slaviae Miltiorrhizae. It has a strong biological activity and has a high medicinal value especially in the fact of the anti-oxidation, free radicals.Salvianolic acid B was transform into Salvianolic acid A by heating process. Transform process was optimizated and the result was evaluated by molar conversion rate. The isolution process was optimizated and the result was evaluated by Yield and purity. The validated process was accepted by pharmaceutical industry scan production. The structure of Salvianolic acid A was identified, and impurities in Pharmaceutical raw materials of Salvianolic acid A were isolated and the structures of the impurities were identified. The research work provide base for quality control of Salvianolic acid A in pharmaceutical industry scan production.The 1% solution of Salvianolic acid B was added by three citric acid sodium and the pH value of the solution was ajusted into 6.0, Salvianolic acid B was transform into Salvianolic acid A in the condition of 125℃and 0.15MPa, for four hours. The chemical reaction product was isolated by chromatographic methed. The preparation process of Salvianolic acid A was optimized according to the result of chromatographic analysis. The structures of Salvianolic acid A and impurity were identified by physicochemical constants and spectrography methods.The transform process of Salvianolic acid A was stable, molar conversion rate reach 50%; the content of Salvianolic acid A is more than 95% by three steps chromatographic isolation. The process was suitable for pharmaceutical industry scan production. The quality of three batches piolit products was confirmed with quality standard of Salvianolic acid A. The physicochemical constants and spectrographic dates of Salvianolic acid A were confirmed with literatures. The impurities, that content more than 0.05%, were identified as Salvianolic acid C and Lithospermic acid. |
