| Salvianolic acid B is the main phenolic acid component in the Chinese traditional medicine Salvia miltiorrhiza Bge. It has shown good prospect in cardiovascular and cerebrovascular disease treatment due to its anti-lipid peroxidation, scavenging free radicals and anti-thrombosis.To offer experimental and theoretical guide for the production and application of Salvianolic acid B, several resins in sale were employed with Salvia miltiorrhiza extract to select the best macroporous resin. Then the static characteristic parameters of the selected resin were investigated; also the dynamic adsorption and desorption technology of Salvianolic acid B in the selected resin was studied to optimize the manipulation conditions.The results showed that AB-8 type macroporous resin is the optimum resin; the initial concentration of the extract has great influence on the adsorption efficiency, and 3.06mg/ml was found to be the best; the adsorption efficiency increases as time extends and attains over 95%in 12 hours; neither high temperature or low temperature favors the adsorption, room temperature is the most feasible; the best ratio of Salvia miltiorrhiza to the macroporous resin is 1 to 2; the desorption rate of Salvianolic acid B increases with the concentration of alcohol rising, and 70%alcohol is sufficient to elute most of Salvianolic acid B.The optimal dynamic conditions were obtained when Salvia miltiorrhiza extract flows through the macroprous resin at the flowing rate of 1.8BV/h for over 12 hours and then Salvianolic acid B absorbed in the resin is washed by 2BV of purified water at the flowing rate of 2.7BV/h,1.5BV of 5% alcohol at 2.7BV/h and 2.5BV of 70% alcohol at 3.6BV/h successively.As high temperature and long time may cause the instability of Salvianolic acid B, the purification should be undergone below 60℃within 4 hours. Besides, Salvianolic acid B proves to be more stable in its alcohol solution than in its aqueous correspondent. |
