Polygonum cuspidatum is a kind of perennial shrubby herb belongs to Polygonaceae, the dried rhizome and root is commonly used traditional Chinese medicine. The main active ingredient Resveratrol from Polygonum is not only a natural anti-toxin, but also a natural antioxidants in plants. The protection of Resveratrol anti-cancer and cardiovascular gradually become a research hotspot in recent years. In this study, the young stems of Polygonum cuspidatum as explants, with its callus induced as test material, by adding the crude extracts of Aspergillus niger, crude protein of Aspergillus niger, crude fat of Aspergillus niger, crude polysaccharide of Aspergillus niger, H2O2, H2O2 quencher and chitosan, the experiment studied that the content of Resveratrol and Polydatin changes in Polygonum cuspidatum callus, and the experiment preliminarily discussed the induction mechanism of H2O2in Polygonum cuspidatum callus. The main of studying results showed as follows:1 The relationship between the crude extracts of Aspergillus niger with the content of RES,PD in the callus of Polygonum cuspidatumIn this experiment, the callus of Polygonum cuspidatum were treated with the crude protein of Aspergillus niger, the crude fat of Aspergillus niger, the crude polysaccharide of Aspergillus niger, the crude extracts of Aspergillus niger.It found that the treatment of the crude protein inhibited the synthesis of Resveratrol, and the treatment of the crude polysaccharide, crude extracts, crude fat are promoted on the synthesis of Resveratrol, which dealt with the crude polysaccharides is higher than the other in the content of Resveratrol. the content of Resveratrol with the treatment of the crude polysaccharides is not only higher than the control, but also higher than the crude extracts of Aspergillus niger; and the content of Polydatin is high by the treatment of the crude fat.2 The relationship between the crude polysaccharides of Aspergillus niger, H2O2, H2O2 quencher with the content of RES,PD in the callus of Polygonum cuspidatumIn this experiment, the callus of Polygonum cuspidatum were treated with the crude polysaccharide of Aspergillus niger, H2O2, H2O2 quencher. It found the crude polysaccharide and H2O2as a elicitor act on the callus of Polygonum cuspidatum, the POD, CAT, SOD activity increased; H2O2 quencher as a elicitor act on the callus of Polygonum cuspidatum, the POD, CAT, SOD activity was reduced.5 Effect of the crude polysaccharide, H2O2, H2O2 quencher on PAL activityIn this experiment, the callus of Polygonum cuspidatum were treated with the crude polysaccharide of Aspergillus niger, H2O2, H2O2 quencher. It found the crude polysaccharide and H2O2 as a elicitor act on the callus of Polygonum cuspidatum, the PAL activity increased; H2O2 quencher as a elicitor act on the callus of Polygonum cuspidatum, the PAL activity was reduced.6 The relationship between the different concentrations of chitosan with the content of RES,PD in the callus of Polygonum cuspidatum In this experiment,we used different concentrations of chitosan Omg/L,50mg/L, 100mg/L,150mg/L, which respectively treated the callus of Polygonum cuspidatum. The results showed that the three concentrations of chitosan promote the synthesis of Resveratrol and Polydatin. And the 50mg/L chitosan is the most obvious.
