Polygonum Cuspidatum is a kind of herbage belongs to polygonaceae. It is a Chinese traditional medicine. The mostly activity component is Resveratrol in Polygonum Cuspidatum. Resveratrol is consider of a promising natural chemical anticarcinogen. The experiment established callus induction with taking stem segment and leaves of as Polygonum Cuspidatum explant, studied extraction process of Resveratrol, the effect on growth states and Resveratrol content by different explant, basic media, light intensity(LI), precursor. The studying results showed as follows:1 Using quadrature experiment studied effect on rate of extraction with solvent, temperature, time as factor. As a result, temperature is the mostly important factor in extraction process, secondly is time, the thirdly is solvent. The best extraction is digestion 48 h with acetidin at 4℃. The content is 13.241μg·g-12 The stem callus growthed fast than the leave callus and the Resveratrol content in stem callus is 5 times than leave callus. The callus of stem and leaves in MS media was highly proliferating than others; its growth rate on FW was significant high at 1% than B5 and N6, is 141.0 %和73.8 % respectively. So the callus in the MS of stem segment was the best.3 Taking leaves and stem segment ofpolygonum cuspidatum as explants, for callus induction and culture in different LI. Growth status of callus were analyzed. Content of Resveratrol was determined by HPLC. The best LI was low light treatment (1340-1560 lx) with prior callus information, growth in vitro proliferation and hardly aging. The resveratrol content was 18.350μg·g-1 max in fresh stem segment weight and 140.074μg·g-1 max in dry stem segment weight, resveratrol content of callus was twice over wild polygonum cuspidatum which gained at the same time.4 The precursor could accelerate the biomass of callus. The biomass is the most with 20 mg·L-1 phenylalanine treatment. Using Richard equation simulation the dynamic state accumulates process. In a certain measure, the precursor adjusts the increase condition. By the growth rate parameter (K), the more phenylalanine concentration, the more callus increasing. The callus under phenylalanine treatment is significant higher at 1% than CK.5 The precursor could raise the activity of Phe zyme. The Phe zyme in callus appear similar change in a growth period. At the 7th day, Phe zyme reach the peak. The more phenylalanine concentration, the high activity increasing, A3 is the best, was 97.0 RU·g-1 Fw more than CK 81.6%. A2 was more than CK 75.6%, A1 was low than CK. After 7 d, activity of Phe zyme in callus was bring down.6 The precursor has no significant effect on soluble protein content. In a growth period, the peak of soluble protein content at 14 d-21 d. 7 The precursor could defer the consenescence in polygonum cuspidatum callus. The long time cultured, the more content of MDA fund in the callus. When content of MDA arrived at the maximum, precursor treatment was significant lower at 5% than CK. The soluble sugar content in callus is increasing by long time, A3>A1>CK>A2.8 The precursor could raise the content of RES in callus. The content of RES is the highest with A3 treatment, is 18. 025μg·g-1FW和241. 489μg·g-1DW. In a growth period, the peak of RES content appears at the 21th day. In a word, the best time to gain the calls is at the 21th day.
