This study dealt with the young stalks of perennial Polygonum Cuspidatum as explants and treating the callus induction as materials. By adding fungal elicitor derived from Aspergillum niger and SA, the experiment studied that the elicitor affected the accumulation of resveratrol in callus. The studying results showed as follows:(1) Screening fungal elicitorsThe fungal elicitors derived from Aspergillum niger,Penicillium sp. and Botrytis cinerea induced Polygonum cuspidatum callus. As a result, they could promote the accumulation of resveratrol, the fungal elicitor derived from Aspergillum niger had the highest inducing efficiency, the amount of resveratrol in treatment was 50.2% higher than that in the check.(2) Effect of the fungal elicitor derived from Aspergillum niger on content of resveratrolThe study screened the concentration,adding time and culture time of the elicitor derived from Aspergillum niger. The result showed that 100 mg/L of the elicitor was added into the medium on the fourteenth day and on the sixth day the amount of resveratrol in Polygonum cuspidatum callus was 2. 25 times higher than that in the check.(3) Effect of SA on content of resveratrolBy screening the concentration,adding time and culture time of SA, the result showed that 7 mg/L of SA was added into the medium on the fourteenth day and on the sixth day the amount of resveratrol in Polygonum cuspidatum callus was nearly 1 times higher than that in the check.(4) Effect of the coordination with elicitor derived from Aspergillum niger,SA,phenylalanine on the content of resveratrolBy adding the different combinations with elicitor derived from Aspergillum niger,SA,phenylalanine, the result showed that they could promote the accumulation of resveratrol and the combination of elicitor derived from Aspergillwn niger-SA-phenylalanine had the highest inducing efficiency.(5)Active composition of the elicitor derived from Aspergillum nigerBy separating the composition of elicitor derived from Aspergillum niger, the result showed that the polysaccharides constituent could greatly promote the accumulation of resveratrol, this treatment was superior to the others. But the protein constituent had slight inhibition on accumulation of resveratrol in callus.(6) Content of ABABy the induction of elicitor derived from Aspergillum niger and determining the content of ABA in callus, the result showed that the content of ABA was a sharp upward trend at beginning, it reached at the peak on the fourth day, and then it was down slowly. But in the check the content of ABA was slow upward trend throughout the entire process. The content of ABA in treatment was higher than that in the check.(7) PAL activityBy the induction of elicitor derived from Aspergillum niger and determining the PAL activity in callus, the result showed that the PAL activity had consistent trend in the treatment and check, they were first decline and after rising. The PAL activity in treatment was higher than that in the check.In the treatment, the PAL activity was a sharp upward trend in the first four days, then it was rapid decline and it reached at the peak on the fourth day. In the check, the PAL activity was slow upward and then down, it reached at the peak on the sixth day.(8) POD activity,CAT activity,SOD activityBy the induction of elicitor derived from Aspergillum niger and determining the POD activity,CAT activity and SOD activity in callus. The result showed that the elicitor derived from Aspergillum niger had some effects on these three enzymes. By determining the POD activity, the result showed that the activity had different trend in the treatment and check, the treatment was down-rising-down-rising-down trend and the check was rising-down-rising trend. By determining the CAT activity, the result showed that CAT activity had consistent trend in the treatment and check, it was rising-down-rising-down trend. By determining the SOD activity, the result showed that the activity had different trend in the treatment and check, the treatment was down-rising-down-rising-down trend and the check was down-rising-down trend.
