Meterials of the study were from six breeds of China (JX, NY, QC, LX, CY, CH), and collecting 1390 genomic DNA was template.Genetic variation of CCK and PP genes were detected by PCR-RFLP, Forced PCR-RFLP, PCR-SSCP and DNA pool sequencing techniques, and association analysis were carried out to assess the effects of genotypes of this genes on growth traits of the Chinese cattle. The objects were to discovery the hereditary characteristics and found molecular markers with significantly functions on important economic traits, and then, they will be to offer genetic information for basis of protects and used of breed resources of Chinese local cattle. The results were follows:1. Detected SNPs and polymorphisms of CCK gene in six breeds of cattleWe deteted the CCK gene in the whole genomic of the six Chinese populations, and discovered 5 novel SNPs. They were scanned 5'area, exton 1, instron and 3'area (NW001494083 617G>A,1224T>C,1275G>C,5200T>C,5340C>Trespectivly, but only 1224T>C,1275G>C located at coding region, and 1275G>C was a missense mutation, the mutation leaded a PVUā…”site disappeared.5200T>C in instron of CCK gene, and the mutation created a Xsp I site. A mutaiton in 5341 C>T of CCK gene, which the mutaiton introduced a EcoR II recognizing site disappeared.617G>A and 1224T>C were detected by Forced PCR-RFLP. More of them were in HWE in study populations (P>0.05). And only the linkage mutation in exon 1 were at moderate polymorphism of NY and LX (0.25<P<0.05), the others all less than 0.25. Genotype distribution x2 tests showed that there wre great difference in different cattle breeds.In exonl, there weren't significantly difference between the NY cattle and LX cattle in genotype distribution (P>0.05); QC and CY, CH have not significantly difference (P>0.05); CY and CH had not significantly difference (P>0.05); JX and CH had significantly difference (P<0.01). In the 5'area, the results showed that JX and NY all significant different with QC, CY and CH (P<0.01), LX and CH had significantly difference (P<0.01), then JX and NY, LX had diversity(0.01<P<0.05), the other breeds had no difference. In 3'UTR, after the anlaysis appeared that NY and QC, CY, CH had significantly difference (P<0.01); QC and JX, LX have significantly difference (P<0.01); JX, LX and CY, CH have difference (0.01<P<0.05); NY and JX, QC and CY have difference (0.01<P<0.05), and the other breeds had no difference.2. Relationships between genetic variation of CCK gene and growth traits in three Chinese cattleThe results of linkage mutiations anlaysed that:At 12 months old in NY cattle, AB genotype of cattles had greater body height than genotype AA individuals (P<0.05); while BB individuals had greater heart girth than the genotype AA carriers (P<0.01). At 18 months old, genotype AB and BB individuals had greater body weight, body length, average daily gain, heart girth than genotype AA (P<0.01); genotype BB individuals had greater body weight and average daily gain than genotype AB cattles (P<0.01). Then at 24 months old, genotype AB and BB cattles had greater body height, heart girth and body length than genotype AA cattles (P<0.01). The other of records had not markedly association (P>0.05). Anlysis JX red cattle, in the 5'area, GA and GG, A A had difference in body depth (P<0.05); GA and AA have difference in siatic end wide (P<0.05); GA and GG, AA have difference in cross department high (P<0.05), GG and AA also have difference (P<0.05); The rest of records had no significant association (P>0.05).Anlysis QC cattle, in the linkage mutiations, AB and AA, BB individuals had difference in hipwidth (P<0.05); AA and AB, BB cattles had difference in kao long and weight. And the 3'area mutiation caused the TT and TC had difference in body depth(P<0.05). The other of data had no significant association (P>0.05).3. The polymorphsim of PP gene and relationships with growth traitsWe used the DNA pool and SSCP methods selected SNPs of PP gene, but had not detected any SNP in the six cattle breeds.
