In this study, flowers and leaves of Tilia miqueliana in different developmental stages as the materials were treated by hot-air drying of 90℃,60℃,30℃and vacuum freeze-drying to investigate the content of water, total falconoid, tea polyphone and amino acid. The results showed that the content of total falconoid in flowers were less than leaves; the content of tea polyphone were more than leaves; the content of amino acid were less than young leaf and more than mature leaf. When the water content is lower 6%, the total contents of total falconoid,tea polyphone and amino acid in mature flowers are the most of flowers of Tilia miqueliana, and the total contents of total falconoid,tea polyphone and amino acid in mature leaves are more than that of young leaves of Tilia miqueliana. To making the total contents of natural active substances maximization, the best drying treatment to mature flowers is vacuum freeze-drying for 200 minutes, while the second-best drying treatment is drying under 90℃for 240 minutes; the best drying treatment to mature leaves is drying under 90℃for 80 minutes, while the second-best drying treatment is drying under 60℃for160 minutes.The supersonic assistance extraction processing of total falconoid from Tilia miqueliana flowers and leaves was studied. By means of orthogonally design with four factors (ratio of solvent to dry Tilia miqueliana flowers and leaves, extraction time, ultrasonic power and ethanol concentration) as influence factors, the optimal extraction technology was performed. As the materials are Tilia miqueliana flowers, the results indicated that the effect of factors on the extraction was in the order as follows, then ethanol concentration, the most important one, extraction time, ratio of solvent to dry Tilia miqueliana flowers, and ultrasonic power as the last one. The optimum conditions were ethanol concentration 60 percent, extraction time 40 min, ratio of solvent to dry Tilia miqueliana flowers 90mL·g-1, and ultrasonic power 80%. With the optimized technology, the content of total falconoid in the final product is 89.51 percent. As the materials are Tilia miqueliana leaves, the results indicated that the effect of factors on the extraction was in the order as follows, then ethanol concentration, the most important one, extraction time, ultrasonic power, and ratio of solvent to dry Tilia miqueliana flowers as the last one. The optimum conditions were ethanol concentration 80 percent, extraction time 40 min, ratio of solvent to dry Tilia miqueliana flowers 30mL·g-1, and ultrasonic power 70%. With the optimized technology, the content of total falconoid in the final product is 88.58 percent.Powders of Tilia miqueliana mature flowers and mature leaves as the materials were prepared total falconoid.The scavenging effect of total falconoid to hydroxyl radical and antioxidant activity were tested. The results showed that the both of Tilia miqueliana mature flowers and mature leaves flavonoid extract had the strong scavenging function to hydroxyl radical; and the scavenging function of mature flowers was greatly stronger than mature leaves,Vc and BHT. The antioxidant activity of Tilia miqueliana mature flowers and mature leaves flavonoid extract were strengthened with the concentration of flavonoids and reaction time.
