In this study, 1062 blood samples from 5 bovine populations Nanyang, Jiaxian Red ,Qinchuan, and Chinese Holstein were collected to extract genomic DNA.Genetic variation of ANGPTL4 and GPIHBP1genes were detected by DNA sequencing, PCR-RFLP and Forced PCR-RFLP techniques, and association analysis were carried out to evaluate the effects of genotypes of candidate genes on growth traits of three Chinese cattle (Nanyang, Jiaxian Red and Qinchuan). The aim were to discovery the Genetic characteristics and to explore molecular markers with significant effects on Growth traits traits for efficient selection and improvement of Chinese cattle, and to provide genetic information for foundation of molecular marker, protection of breed resource of Chinese bovine.The results were as follows:1. The bioinformatics analysis of bovine ANGPTL4 geneThe analysis of amino acid sequence indicated that cattle ANGPTL4 gene consisted 7071 bp,7 exon and 6 inexon,coded 410 amino acids. Homologous comparison indicatedcattle ANGPTL4 gene similarity in nucleic acid sequence with pig, the genetic relationship is closer .Bovine ANGPTL4 gene cDNA was found in liver and Adipose tissues by the analysis of its expression in various tissues.2. SNPs detection of ANGPTL4 gene and their correlation Analysis between growth traits in Nanyang, Jiaxian Red and Qinchuan cattle.SNPs were detected in the exon from 5 cattle breeds in China. Comparison between the nucleotide sequences of the bovine ANGPTL4 gene and the above sequences revealed 4 SNPs of the ANGPTL4 gene. The 1422T>C was missense mutations, While variation at the other 3 SNPs were synonymous mutations.In Nanyang cattle population, different genotypes were striking associated with the body weight and Average daily gain of 12 month, Hucklebone width of 18 month, which the mutational individuals were greater than wildtypes (P<0.05). In Jiaxian Red cattle population, different genotypes were striking associated with the hip width and Chest width of which the mutational individuals were greater than wildtypes. In Qinchuan cattle population, different genotypes were striking associated with the height at hip cross and hucklebone width, of which the mutational individuals were greater than wildtypes (P<0.05). 3. SNPs detection of GPIHBP1 genePCR reactions were carried out with the mixed DNA, and then the products were directly sequenced, whose results were compared with the sequence supplied by the GenBank (NC-00732.4). It showed two mutations within the 896 bp fragment:the base transversion of G>C and the base transition of C>T were respectively located at 309 bp 319 bp within intron of bovine GPIHBP1 gene and the distance between G309C and C319T was only 10 bp. There wasn't mutation within the 664 bp fragment for exon 3 of bovine GPIHBP1 gene. A base transition of C>T was detected within the 445 bp fragment for exon 4, located at 1762 bp of coding region, which caused the codon GGC was changed to GGU, belong to the same mutation of Gly.
