| Angiopoietin like protein 4 (Angiopoietin-like protein 4, ANGPTL4) class of an giogenesis) gene is secretory protein which is related to fat metabolism. And ANG PTL4 gene not only regulates fat and energy metabolism, but also participate in the process of cell proliferation, apoptosis regulation and muscle growth regulation. Th us. it plays an important role in the process of Muscle growth and development. T his experiment selected Tianfu black rabbit, IRA rabbit and New Zealand rabbit as experimental materials, using PCR-DS to detect SNPs of ANGPTL4 gene coding re gion, analysing linkage equilibrium, using PCR-RFLP technique to proceed genetic t yping in genetic mutation locus, and doing a correlation analysis on SNPs and gro wth trait of rabbits. The main results are as follows:1.After comparing growth traits of TianFu black rabbit, YiLa rabbit and New Zeala nd rabbit at the age of 35,70 and 84 days, it shows that there is no obvious diffe rence between them, but according to multiple comparison, the weight of Tianfu bla ck rabbit at the age of 70days and 84days is higher than IRA rabbit(P<0.05); the weight of New Zealand rabbit is obviously higher than IRA rabbit at the age of 28 days and higher than other breeds at the age of 35days; the weight of New Zeala nd rabbit is higher than IRA’s at the age of 70 and 84 days.2.There is no obvious difference between average daily gain weight of different rabbits at the age of 28~84 days, but average daily gain weight of Tianfu black r abbit is higher than New Zealand rabbit and New Zealand rabbit is higher than IR A rabbit.Three SNPs are detected in the ANGPTL4 gene exon 6. One of them is missense mutation, the others are synonymous mutation, respectively c.827 G>A、c. 867 T>C and c. 875 T>C Through predicting protein domain by SMART online a nalysis software, it is found that the three locus are located at FBG structure. Then it shows that c.827 G>A is highly chained with the other two SNPs (r2≥0.75). Th us a conclusion can be reached that this SNPs have a similar genetic effect to othe r two SNPs, and finally c.827 G>A is chosen to proceed subsequent research of ge netic polymorphism.3. AA, AG and GG three genes are detected in the c.827 G>A of ANGPTL4 gene in three rabbit breeds, and all are moderate polymophiclocus(0.25<PIC<0.50); χ2 examination shows that c.827 G>A is in equilibrium in the three rabbit breeds.4. The correlation analysis of c.827 G>A SNPs in ANGPTL4 gene and growth trait shows that among the three rabbit breeds, hereditary effects of average daily gain weight at the age of 70,84 days and 28-84 days reach significant level(P<0.05); average daily gain weight of GG gene is obviously higher than AA gene(P<0.05) at the age of 70,84 days and 28~84 days and higher than AG gene; the weight of GG gene is higher than AA and AG gene at the age of 28 and 35 days. |
