A.sinensis as a perennial medicinal plant is a part of umbelliferae angelica linn.The root of A. sinensis very good for enrich the blood, regulating menstruation and relieve the pain, laxation intestines and so many virlue,it also use to cure disregulating menst-Ruation, blood deficiency and dizziness, traumatic injury, rheumatism and so on.The height of Gansu noth min hill is 2 000-2 800m, A. sinensis is cultivate in these wet and alpine mountainous area, for example Min county, Zhang county, Weiyuan county and so on. Every year the average of cultivate area is 20 000 hectare, annual output about 6 500t.The cultivate area of A.sinensis in Gansu take up seventy percent though out the country, occupy the 90 % above that A.sinensis total output completely, Become a pillar industry for economic development, it is good for poverty areas overcome poverty and achieve prosperity. But the bolting restrict A.sinensis cultivate, ordinary the incidence of A.sinensis bolting is 30%~50%. Sometime the bolting rate will achieve 90%.the bolting A.sinensis will lose medicinal value, because the root of bolting A.sinensis gradually became lignification, the bolting is very harmful for A.sinensis cultivate.This study by determine glycometabolism and the active of POD,PPO and so on, to know about the mechanisms of the protein and enzyme metabolic in the bolting A. sinensis, giving clarification the process of bolting A.sinensis physiological and biochemical changes. Provides the theory basis for prevent A.sinensis bolting.The results show that there were some difference between bolting and unbolting A.sinensis in the same period. Ten day before the floral bud, The bolting A.sinensis content of free amino acid,POD and PPO active were 7.53 mg/(gFW),0.097ΔA470/ (min·gFW),0.087ΔA410/(min·gFW), higher then the bolting plant 1.3 mg/(g·FW),0.02ΔA470/ (min·gFW),0.014ΔA410/(min·gFW). The content of soluble sugar and soluble protein in bolting plant were 4.63% and 0.94% ,higher then the bolting A. sinensis 2.2% and 0.5% . In bolting stages, the content of soluble sugar and soluble protein were higher in unbolting A. sinensis, the content of free amino acid,POD,PPO were higher in bolting A. sinensis. So can be conclude that the A. sinensis bolting is very compecate physiological reaction and control by lot of metabolites. By use different method extract total RNA of the bolting and unbolting A.sinensis, using DDRT-PCR technique anlysis bolting A.sinensis different diplay gene. Optimize every parametric that influence PCR amplify system of the DDRT-PCR technique, ultimate make sure the PCR amplify system and PCR amplify procedure that fit for A. sinensis DDRT-PCR study. Blast the eight different gene from the PAGE found these gene is very similar with Berberis bealei 18S ribosomal RNA gene and Viola philippica 18S ribosomal RNA partial sequence, B.rapa chloroplast petA gene for cytochrome f, the mRNA of Arabidopsis lyrata subsp lyrata cbl-interacting protein kinase 7 (CIPK7) gene and Arabidopsis thaliana Full-length cDNA Complete sequence from clone GSLTFB95ZF12 of Flowers and buds of strain col-0 of Arabidopsis thaliana. But if these gene connect with the A.sinensis bolting, need to move forward clone the whole cDNA of bolting A.sinensis and analyse its function.
