In this paper,the leaves,roots and petioles of Angelica were used as materials to measure the activities of indole acetic acid oxidase, amylase and amylophosphorylase as well as soluble protein content, at different parts and different periods;through the preparation of hormones artificial antigen and the establishment of indirect enzyme-linked immunosorbent assay (ELISA) method, the changes of plant endogenous hormone (IAA and ABA) content in Angelica were measured. The results revealed the inherent factors in the process of bolting and contributed to the solution of early bolting. The main research results are as follows:In the preparation of Hormone Polyclonal antibody, EDC and mixed acid anhydride method were applied to produce artificial antigen of IAA and ABA respectively. The artificial antigen was tested by uv spectrophotometry and SDS polyacrylamide electrophoresis to insure the quality. A mouse was used as model to generate Hormone Polyclonal antibody.According to the principle of indirect ELISA, appropriate reaction condition of ELISA was established in order to measure the content of IAA and ABA at different parts and periods. The results demonstrated that the content of IAA in leaves and petioles of bolting was apparently higher than unbolting plants which indicated the positive of high concentration IAA in blossom. The content of ABA in leaves, petioles and roots of bolting was higer than unbolting plants which indicted that high concentration ABA would benefit blotting.The activity of indole acetic acid oxidase, amylase and amylophosphorylase were tested and the results indicated that the oxydase activity of unbolting plants was higher than bolting in Angelica; the higer the oxydase activity was, the lower the content of IAA would be. The result may due to the positive role of IAA in blottiong.The activity of amylase and amylophosphorylase in bolting was higer than unbolting plant in Angelica. The reason may be that the energy was provided by amylolysis in the process of bolting to satisfy the demand of blossom. Bradford method was applied to measure the content of soluble protein and the results demonstrated that the content in bolting was lower than unbolting. The reason may be that the accumulation of soluble protein would benefit bolting of Angelica. The bolting of Angelica consumed large amount of nutrition so that the soluble protein was lower than unbolting plants.
