Studies On Propagation Technique Of Tissue Culture And Cutting Of Two Varieties Of Ornamental Clematis

Clematis has colorful blossoms and beautiful showy. So it is a valuable ornamental plant.In this experiment, tissue culture and cutting propagation of Clematis 'Vyvyan Pennell' andClematis 'Andromeda' were studied.The main results were as follows:1. The study of tissue culture technology(1) The results of sterilization of explant: Stems with buds and stems were used as explants.Thebest sterilization time by HgCl₂(0.1%) was5minutes to the stems with buds,3minutes tothe stems of Clematis'Vyvyan Pennell',8minutes to the stems of Clematis'Andromeda'.(2) Initial culture of stems with buds: The best initial medium was MS+1mg/L6-BA+0.05mg/L NAA, The germination rate of Clematis 'Vyvyan Pennell' was100%, Thegermination rate of Clematis'Andromeda'was92.86%.(3) Multiplication culture: The best culture medium for Clematis'Vyvyan Pennell' buds prolif-eration was MS+2mg/L6-BA+0.02mg/L NAA. The proliferation index was2.75. ToClematis 'Andromeda'was MS+0.05mg/LTDZ+0.02mg/L6-BA. The proliferation indexwas2.62.(4) Rooting culture: The best medium for rooting of Clematis'Vyvyan Pennell'was1/2MS+1mg/L NAA+0.5mg/L IBA and the rooting rate was50%.(5) Callus induction of stems: The medium of MS+1mg/L6-BA+1mg/L NAA and MS+0.125mg/L TDZ+1mg/L2,4-D both can induct callus. The callus induction rate was above90%.2. The study of cutting propagation technology(1)Treatment of plant growth regulator: Half of woodiness branches of Clematis'Andromeda'were used as materials.The control group had the highest rooting rate which was up to53.56%.(2) Matrix treatment: Half of woodiness branches of two varieties of Clematis were used asmaterials. To Clematis'Andromeda',the best cutting matrix was in the perlite+vermiculite+peat which ratio was1:1:1.The rooting rate was35.71%.To Clematis'Vyvyan Pennell'was in the vermiculite+peat which ratio was1:1. The rooting rate was66.67%.(3) Relationship of rooting and enzymes activity: Using plant growth regulator accelerated thechanges the activity of POD,SOD,PPO and took rooting in advance.
