As one of the most important economic crop and extensive cultivated vegetables, Chinese cabbage (Brassica campestris L.ssp.Pekinensis) were originated from China. Chinese cabbage are cultivated all over the world. But the tipburn in Chinese cabbage is one of the most serious diseases. It is a physiological disorder induced by calcium deficiency.Different variety has significantly difference in resistance to tipburn.Therefore, breeding the resistant variety to tipburn disorder is the most economic and efficient method.The traditional breeding methods is the cost in time and effort and the result is not accurate enough. With the development of molecular biology techniques, molecular markers related to tipburn resistance are screened, which has great significance for breeding. In this paper, we studied and contrasted different identifying methods of tip-burn and choose resistance lines "xiaobaoC8",susceptible lines "651A" and F2 as materials, and the AFLP markers were employed in screening molecular markers related to tipburn gene in Chinese cabbage. Results as following:1 We studied different identifying methods of tip-burn,the results show that:the method of solution culture has high accuracy and typical symptom, which can be used to identify of tip-burn effectively.2 Through identifying of tipburn of F2 population, we acquired 53 resistant plants and 83 susceptible plants among 136 F2 plants.3 Molecular markers of tipburn genes were screened by AFLP combination with BSA, E01/M01 prime combination was used in preamplification and E+3/M+3 prime combination was used in amplification. We obtained one polymorphic bands in xiaobaoC8,651A,resistance pool (BR) and susceptible pool (BS) with 210 primer combinations. We named the polymorphic bands E10/M05.4 The correlation coefficient between the marker E10/M05 and the disease index is 0.633, the marker is related to tipburn genes.5 The sequences were analyzed in the NCBI database and did not search for homologous sequences.
