Breeding And Utilization Of The Genetic Male Sterile Line In Purple Stalk Chinese Cabbage

In order to resolve the problem of cross breeding in utilizing hybrid vigor in Brassica campestris L. ssp. parpurea, according to the genetic characteristics of sterility, the experiment was designed by using the material of multiple allele inherited Chinese cabbage as the source of male sterility. The male sterile multiple allele was transferrd to the sterile inbred line XA500, to chose the nuclear male sterile line of purple stalk Chinese cabbage, whose characters were close to line to be transferred. The main results were as follows.1. Directional transfer scheme was progressed for the purpose of transferring the male sterile gene to the selected fine lines of purple stalk Chinese cabbage whose genotype is MsfMsf and breeding GMS lines which had stable inheritance and were similar to the male fertile lines in botany and economic characters. A stable genetic male sterile line GMS500 'AB'line and'temporary maintainer line'of purple stalk Chinese cabbage were obtained after three consecutive generation of backcrossing. Fertility segregation ratio of each generation was consistent with the theory during transferred expriment, its was proved that the transferring pattern of nuclear male sterile line was feasible in purple stalk Chinese cabbage.2. The fertility identification results of nuclear male sterile line GMS500 of BC3 showed that its stamens were completely degenerated and degree of sterile plants and sterile flower reached to 100%. Comparison of horticulture character between nuclear male sterile line GMS500 and lines XA500 to be transferred showed that characters of GMS500 were very close to XA500 expect from stamen fertility, breeding goal was achieved.3. In order to determine the combining ability of GMS, hybrid combinations was prepared with the male sterile line of BC3 and four inbred lines,'Ziting 11'as the control, a excellent combination was obtained,which had quality traits and its yield was higher than the control, cultivated in greenhouse in Shenyang in autumn, from sowing to harvest about 60 d.
