Triptolide (Tripterygium wilfordii Hook.f.) of Euonymus concores public vine of the genus plants,widely distributed in southern China provinces. The dried root is a traditional Chinese herbal medicine,Tripterygium wilfordii and its preparation have good response to many kinds of disease.Tripterygium newdrug has caused extensive attention in medicine, its application field is constantly expanding, and therelevant demand is getting more and more with a very broad application foregroun. But the TripterygiumWilfordii is definite toxicity, so study on Tripterygium wilfordii has also become a focus of attention andhot research topic. Because of its extensive sources, medicine Tripterygium wilfordii habitat difference isbig, the production process caused by the different material quality uneven; it's quality control is lack ofsystematic study. All these factors can cause Chinese herbal medicine and its pharmaceutical quality differgreatly and it has seriously affected the safety and effectiveness in the clinical applicaton.The article study on Tripterygium wilfordii effective components and fingerprints of32provenancesfrom8provinces,26cities. Established the optimum extraction conditions of the effective component;initialestablished the relationship between factors and quality control of Tripterygium wilfordii;obtained thefingerprint of different provenances. The main results are as follows:1. Triptolide axtrxaction with the help of four two hand in and rotate and design, obtained the optimalextractio: methanol concentration78.64%material/liquid1:35.96, ultrasonic time39.1min, chromatographyalumina content9g, can obtain the maximum triptolide amount for0.0192%.2. Analysis the root and leaf of32provenances, it was found that Tripterygium wilfordii root and leaftriptolide content were the highest from Fujian Taining, the highest leaf triptolide content from SanmingTaining, the content reaches26.37μg·g-1;the highest root triptolide content from Sanming Datian, thecontent reaches26.37μg·g-1.Tripterygium wilfordii root and leaf triptolide content were the highest fromHunan Xiangxiang, leaf triptolide content reaches8.94μg·g-1, root triptolide content reaches7.73μg·g-1.3. In the same plant the same parts in different seasons of the secondary metabolites contents isdifferent..Leaf and branch triptolide content increased from January to August and reached the highestvalue, as the metabolism and the weakening of tetrandrine as temperature decreases, decreased to aminimum by February. The root of a peptide has been accumulated from March to January the next year, atthe end of January the root of a hormone content reaches highest, with the temperature and plant growthand metabolism, the root of a hormone decreased dramatically, in the third year reaches a minimum value.4. In different parts of the plant triptolide content is different. Branches of triptolide content was thelowest, and compared the branches and limbs of the triptolide content, it was found that main branches andside branch triptolide content showed no significant difference. Leaf pigment is about23times shoots,slightly higher than the root content.5. Triptolide content accumulation with growth years grow, but more than a certain number of years,triptolide accumulation will decline.5years triptolide content is highest,6years triptolide slightly lower,therefore suggested that5years triptolide should be updated. 6. Triptolide content was highest when the drying temperature is60℃,30.26μg·g-1the minimumtemperature is100℃,10.11μg·g-1in order to ensure the quality of Tripterygium wilfordii, the dryingtemperature should be controled within6065℃.干燥温度是60℃根部甲素含量最高,达30.26μg·g-1,100℃含量最低,仅10.11μg·g-1。7. Tripterygium wilfordii fingerprint to establish the optimal chromatographic conditions are asfollows: wavelength267nm, flow rate1.0ml min-1, column temperature40℃, the mobile phase: A-1%phosphoric acid, B-acetonitrile, elution gradient of010min,85%65%B;1030min,65%50%B;3055min,50%40%B;55;70min,40%30%B;7090min,30%15%B;90100min,15%10%B.8. In the elevation of the differences will also affect the quality of Chinese medicinal materials. Thehighest content of test materials from the elevation of420m, leaf triptolide content reaches a maximumvalue of24.33μg g-1.9. Tripterygium wilfordii fingerprint in21common peaks, the common peaks relative retention timeand relative peak area reproducibility and precision of RSD <3%, shows that the method can be used forproduction of Tripterygium wilfordii fingerprint chromatogram.10. According to different sources of Tripterygium of the common peaks in fingerprints and therelative retention time and relative peak area undertake clustering, clustering results differ essentiallysimilar, different sources are divided into four categories, a source similarity and geographic distance andlittle correlation.
