Xinjiang is one of the country's main agricultural and pastoral areas.over the lastdecade, because of the over-grazing and natural disasters, natural grasslanddesertification areas are constantly increasing. So utilization ofcorn,wheat,alfalfa,sorghum,cotton are very important in the desertification problemdealing with. lactic acid bacteria plays very critical role in silage making by usingthose straw resources which are the main basic material in traditional silageindustrges. In this researched classification and diversity of the identification lacticacid bacteria resources isolated from the crops such ascorn,wheat,alfalfa,sorghum,cotton in Turpan of xinjiang were studied respectively.The lactic acid bacteria were isolated from the five kinds of forage inxinjiang.by using plate separation method(19strains of lactic acid bacteria wereobtained from alfalfa,18strains from corn,45strains from straw,14strains fromsorghum,7strains from cotton). These63strains of lactic acid baceterias is Coccillus,40strains is Bacillus. The morphological,physiological and biochemicalidentification showed that the103strains of lactic acid bacteria belonged to twogenera,namely, Lactobacillus and Enterococcus.40strains of Which belong to thegenus lactobacillus, accounting for38.83%, including lactobacillus casei,lactobacillus rhamnosus,lactobacillus paracasei, lactobacillus plantarum;63strainsbelonging to Enterococcus, accounting for61.17%, including Entercoccus faecium,Entercoccus durans, Entercoccus hirae. for the separation of hingh quality lactic acidbacteria using for high quality silage production, among84stains selected32strainslactic acid becteria,which were6strains from cotton,14strains from sorhum,6strainsfrom feed corn straw,6strains from weat strawof better acid-production ability wereselected to identified salt tolerance,acid tolerance and the sequence analysis of16SrDNA. Identification results have been selected out of32strains belong to two genera, lactobacillus, Enterococcus. The32strains show better salt-tolerance andacid-tolerance ability.16S rDNA sequence made BLAST contrast with internationalnucleic acid database GeneBank,to classify species,at the samae time,usingCLUSTAL-W software blasted the related species,using MEGA4.0software toconstruct phylogenetic tree. The result of16SrDNA identification:084,080,028,024,023,087,091,086,079,082,081,083,088,0101,055,0110,058,095,0103,066,02,092,020were Entercoccus faecium,0104were Entercoccushirae,018,0111,0108,097,033,019were lactobacillus plantarum,0105werelactobacillus casei. Identification results have been selected out of Thirty-towbelong to two genera, Lactobacillus, Enterococcus. The Thirty-tow strains showbetter salt-tolerance and acid-tolerance ability.16SrDNA sequence made BLASTcontrast with international nucleic acid database GeneBank,to classify species,at thesamae time,using CLUSTAL-W software blasted the related species,using MEGA4.0software to construct phylogenetic tree. In this study,4strains Lactic acid bacterias with batteracid production ability, to provide a theoretical foundation and scientific basis theresources exploitation of lactic acid bacteria in five forage from Turpan of Xinjiangand production of high quality silage.
