Potassium is an essential element of the growth of tobacco, and potassium content is an important indicator of the evaluation of the quality of tobacco, it has an important role in improving the tobacco leaf maturity, aroma and taste, flammability, and security. Potassium content in China's tobacco is generally low, improve the tobacco potassium content can increase the competitiveness of China's tobacco industry.In this study, the Strains of high potassium New Tobacco GK5as the male parent to hybridization the china tobacco86as the female parent.Analysis of its main traits advantage of the characteristics; Using SRAP markers to identified potassium gene,at the molecular level research differences in potassium content between the different flue-cured tobacco.The results showed that:1.The average of hybrid generation (F1) potassium content is2.65%,higher than the female parent(2.11%) ratio is11/24; higher than the male parent(3.24%) ratio is3/24.The average of hybrid second generation (F2)potassium content is2.73%, higher than the china tobacco86ratio is47/50, higher than the GK5ratio is17/150.The ratio of the potassium content is higher than or equal to2.69%in the F1generation is11/24,the ratio of the potassium content is higher than or equal to2.69%in the F2generation is8/15.2. The advantages of ultra-high affinity and ultra-low affinity of F1generation are not obvious in the fertility of the performance,respectively-0.057,0.009.The advantages of ultra-high affinity of F1generation is-2.00in the natural incidence,the advantages of ultra-low affinity is0.53. Ultra-high affinity advantage of F1generation of potassium is-0.59, ultra-low affinity advantage is0.26.In the agronomic traits, the plant height average advantage of is0.323that is the highest in the F1generation, the second highest is leaf length0.059, the leaf width, girth of stem, leaf number, pitch, respectively-0.006,-0.010,-0.008,-0.001.3.Screening for100pairs of upper and lower primers, which six pairs of primers could amplify specific band among the parental, F1, F2, and control, potassium content higher than2.69%can be detected a specific band, and less than2.69%of the material can not be detected specific band. SRAP markers can identification of differences on the molecular level between difference level of potassium content of flue-cured tobacco.
