| Objective:Endometriosis(referred to as endometriosis,EM,endometriosis)is the common disease and frequent disease of reproductive women.It is benign,but has malignant biological behavior of infiltration,metastasis et al.Because of its obvious clinical symptoms.and the face that it's so easy to recurrent after treatment,so it got more and more attention.Now surgical treatment is the main treatment method,but because of the high recurrence rate,the patients'prognosis is not so optimistic.So surgical combined drug treatment is the main direction.At present,drug assistance treatment make ectopic endometrium lesions atrophy by reducing the level of estrogen,to obtain the purposes of controlling disease's development and reducing the clinical symptoms.However osteoporosis, weight gain and other side effects effect the patient's compliance,and the recurrence rate is still not improved. In this experiment, we took the SD rats as experimental subjects,established endometriosis rats model by autologous transplantation.Then we observed the change of the level of Turner necrosis factor-alpha(TNF-a) after the treatment used cisplatin by peritoneal lavage to investigate the effect and mechanism,then to provide new ideas and methods of treatment and the basis for its clinical application.Content:We choosed SD rats as experimental subjects,established endometriosis rats model by autologous transplantation.The Cisplatin were used as therapy drug to treat edometriosis by peritoneal lavaget.And made physiological saline treating group and model control group as contrast.Took the level of TNF-a in the peritoneal fluid as index.to reflect the treatment effect of Cisplatin by peritoneal lavage according to the changes of content.Method:Used of clean, healthy, female, no mating SD rats (45) as experimental subjects.They were divided randomly to two groups.One of them was endometriosis model group,the rats in this group were used to established endometriosis rats model by autologous transplantation.The other group is normal control group,the rats in this group were recepted sham-opration by stretching uterine only after opening the abdominal cavity.4 weeks after surgical,we opened the abdominal cavity again,observed the growth of implanted endometrial and collected samples. EM rats were divided randomly into three groups that were deal with different methods.Cisplatin therapy group were given Cisplatin 2mg/kg+distilled water2ml by peritoneal lavage. Physiological saline treating group were given a large number of normal saline (10ml) by peritoneal washing.The model control group were not given treatment after collected samples.4 weeks later,opened the abdominal cavity,observed the growth of implanted endometrial and collected peritoneal fluid samples.Then we detected the content of TNF-a in the peritonral fluid of rats of each group by Enzyme Linked-Immuno-Sorbent Assay(ELISA),to observe the changes and made statistical analysis.Part of ectopic lesions were taken to make histopathological lesions detection in every surgery.Result:EM rats model were established successfully by confirmed of histopathological lesions detection.The level of TNF-a in peritoneal fluid of EM rats were significantly higher than the level of the normal rats(P<0.05).After different treatment, the level of TNF-a in peritoneal fluid of Cisplatin therapy group were significantly lower than the level of the physiological saline treating group and the model control group(P<0.05).The level of TNF-a in peritoneal fluid between the latter tow groups had no significant different(P>0.05).Conclusion:Autologous transplantation can establish EM rats model. The level of TNF-a in peritoneal fluid of EM rats were significantly higher than the level of the normal rats, TNF-a may play a certain role in the occurrence and development of endometriosis.Cisplatin can reduce the level of TNF-a in peritoneal fluid by peritoneal lavage,and the treatment may expected to be the assistant therapy of the surgery to reduce the recurrence rate and enhance the results of operations. |
