Objective:To investigate the expression level of three mRNA(Shh, Smo and Glil) of Hedgehog(Hh) signaling pathway in human hepatocellualr carcinoma(HCC) and evaluate its clinical significance in the hepatocellular carcinogenesis.Methods:Reverse transcription polymerase chain reactions (RT-PCR) was performed on 20 samples of hepatocellualr carcinoma from the First Hospital Affiliated to Nanchang university(2009.4-2010.4)and their corresponding tumor-adjacent tissues and 10 samples of normal liver tissues in the same period for the expression levelof three mRNA (Shh, Smo and Glil) of Hh signaling pathway. The relationship between the expression level of three mRNA in HCC tissues and related clinical parameters was analyzed.Results:(1) The expression level of the three mRNA (Shh, Smo and Glil) of Hh signaling pathway in the HCC tissues were all markedly higher than in their corresponding tumor-adjacent tissues and normal liver tissues (Shh mRNA: 0.594±0.113 Vs 0.461±0.128,0.594±0.113 Vs 0.459±0.081 P<0.05; Smo mRNA: 0.595±0.107 Vs 0.481±0.104,0.595±0.107 Vs 0.439±0.102 P<0.05; Glil mRNA: 0.551±0.142 Vs 0.442±0.104,0.551±0.142 Vs 0.435±0.094 P<0.05).(2)There was a positive correlation between the expressions of Shh mRNA and Smo mRNA in HCC tissues (r=0.740, P=0.001). There was also positive correlation between the expressions of Shh mRNA and Smo mRNA in their corresponding tumor-adjacent tissues(r=0.680, P=0.001) and normal liver tissues(r=0.760 P=0.011). Also the expressions of between Shh mRNA and Glil mRNA was a positive correlation in HCC tissues(r=0.582,.P=0.007), and the same results in their corresponding tumor-adjacent tissues(r=0.688, P=0.001) and normal liver tissues(r= 0.772, P=0.009). Last, of between Smo mRNA and Glil mRNA, the expressions were all positive correlation in HCC tissues(r=0.559, P=0.010), their corresponding tumor-adjacent tissues(r=0.534, P=0.015) and normal liver tissues (r= 0.683 P=0.029).(3)The expression of three mRNA(Shh, Smo and Glil) of Hh signaling pathway in the HCC tissues with the pylic cancer embolus was obviously higher than that of with no pylic cancer embolus (Shh mRNA:0.667±0.054 Vs 0.523±0.090 P<0.05; Smo mRNA:0.658±0.063 Vs 0.544±0.110 P<0.05; Glil mRNA:0.650±0.077 Vs 0.470±0.132 P<0.05). Also, the expression of the three mRNA(Shh, Smo and Glil)of Hh signaling pathway in the HCC tissues that the maximum diameter in tumor is equal or greater than 5cm is obviously higher than that of their maximum diameter of tumor less than 5cm(Shh mRNA:0.488±0.097 Vs 0.631±0.076 P<0.05; Smo mRNA: 0.476±0.066 Vs 0.646±0.075 P<0.05;Glil mRNA:0.402±0.060 Vs 0.615±0.116 P<0.05).Conclusion:(1)The expression level of the three mRNA(Shh,Smo and Glil) of Hh signaling pathway in the HCC tissue were all markedly higher than in their corresponding tumor-adjacent tissues and normal liver tissues, this showed that the Hh signaling pathway have closed relation with HCC and further Speculated that Hh signaling pathways played an important role in the occurrence and development of HCC.(2) There was a significantly positive correlation and synchronicities between the expressions of Shh, Smo and Glil mRNA in HCC tissues, and also in their corresponding tumor-adjacent tissues and normal liver tissue. This phenomenon had Accorded with the Chain reaction rule of Hh signaling pathways in the occurrence and development process of HCC.(3) the expression of the three mRNA(Shh, Smo and Glil) of Hh signaling pathway in the HCC tissue that tumor is bigger and with the pylic cancer embolus is obviously higher. This showed that the role of Hh signaling pathways played in the whole development process was strengthened with the development of HCC, and further explained that Hh signaling pathways may participate in the invaded and distant metastasis of HCC.
