During the chemical investigations on this medicinal plant(Cornus officinalis), 31 compounds including 4 new ones were isolated and identified. The dissertation is composed of two chapters.The research progresses of chemical constituents and pharmacological activities of C. officinalis, which were reviewed in the first chapter.The chemical constitutes of C. officinalis were discussed in the second chapter, this plant is a member of the Cornaceae family. This herb (Fructus corni), known as Shan-zhu-yu in Chinese, has been used to treat cerebrovascular disease and diabetes in Traditional Chinese Medicine for a long time. However, so far, the chemical constituents of this species distributed in Anhui Province of China have not been reported. To compare the chemical constituents'differences of the same Cornus species belonging to different geographical distribution and climatic conditions, we investigated the fruits of C. officinalis collected in Shitai region of Anhui Province, China. In this work,31 compounds were isolated from the methanol extract of the fruits of C. officinalis, mainly including iridoids, sterins, flavonoids, polyhydric alcohol, phenols and phenolic glycoside, and pentacyclic triterpenoide. They were ursolic acid (1),β-sitosterol (2), daucosterol (3), cornuside (4),7-α-O-methylmorroniside (5), 7-β-O-methylmorroni- side (6), loganin (7),7-α-morroniside (8),7-β-morroniside (9), secoxyloganin (10), caffeic acid (11), quercetin-3-(9-β-D-glucuronide methyl ester (12), quercetin-3-O-β-D-glucopyranoside (13), quercetin-3-O-β-D-galactopyranoside (14),2-O-(4-hydroxybenzoyl)-2,4,6-trihydroxyphenyl-methylacetate (15), kaem-pferol-3-O-β-D-galactopyranoside (16), quercetin (17), rutin (18), quercetin-3-O-α-L-rhamnosyl-(1â†'6)-β-D-galactopyranoside (19), kaempferol-3-0-α-L-rhamnosyl-(1â†'6)-β-D-glucopyranoside (20), (2S,3R,4S,5S)-2-(hydroxyl-methyl)-2-methoxytetrahydro-2H-pyran-3,4,5-triol (21),7-a-O-ethylmorroniside (22),7-β-O-ethylmorroniside (23), cornusideâ…¢(24), cornusideâ…£(25), methyl quinate (26), loganic acid (27), tachioside (28), (-)-Isolariciresinol-9'-β-glucopyranoside(29), (1'S,2'R)-Guaiacyl glycerol 3'-O-P-D-glucopyranoside (30), mevaloside(31). Among them, compounds 15,21,24,25 are new ones,10-16,18-21,24-26,28-30 were isolated from this plant for the first time. Their structures were established by spectroscopic analysis, especially using 2D-NMR techniques (1H-1H COSY, HMQC, and HMBC) and comparisons of their data with literature values. The compound of 15 and 31 were further confirmed by X-ray analysis.
