Objective:Discussion on response of mouse asthma against inhalation of naphthalen with different concentrations.Methods:Selected 48 BALB/c of mice 6 to 8 weeks age, were randomly divided into 6 groups, in each group 8 mice (n=8):A group (control group), B group (ethanol group), C group (asthma model group), D group (naphthalene low concentration group), E group (middle concentration group), F group (high concentration group). In C group, using freshly prepared 20μg ova albumin(OVA)+1 mg aluminum hydroxide intraperitoneal on the 1,8,15 days sensitized, on 22 day started OVA (10mg/ml) inhalation 30 minutes,1 time every day for 7 days. In A, B groups, normal saline aerosol inhalation used instead of intraperitoneal ovalbumin injection respectively. D, E, F groups were given OVA too, Step with the asthma model group, after the inhalation by Asthma model of low concentrations (50mg/m3), middle concentration (250mg/m3), high concentrations (500mg/m3) naphthalene respectively, exposed to smoke in the glass box for acting gas exposure,4 hours a day, for 2 weeks. Meanwhile A, B and C group also same size glass box in receiving aerosol inhalation,A group of aerosol inhalation with normal saline,B,C group aerosol inhalation with ethanol,4 hours a day for 2 weeks.1 week after the end of naphthalene, inhalation OVA (10mg/ml) inhalation given for 30 minutes. Evaluate RL of each rat; by enzyme-linked immunosorbent assay (ELISA), determination of serum IL-4,IgE changes; determination of lung tissue superoxide dismutase (SOD), glutathione peroxidase (GSH-PX) activity and malondialdehyde (MDA) content changes; in lung tissue sections were stained with HE,pathological changes of lung tissue observed.Results:①airway hyperresponsiveness did not occur in mice in A, B groups, in the C,D, E, F groups, airnificantly significantly increased compared to C group, the difference was statistically significant (P<0.01), D, E, F group with the naphthalene concentration RL of the increaser, the difference was statistically significant (P<0.01).②C, D, E, F the serum concentration of IL-4 than the A, B were significantly increased, the difference was statistically significant (P<0.01), C, D, E, F between the two groups no significant difference (P> 0.05).③C, D, E, F the serum concentration of IgE than the A, B were significantly increased, the difference was statistically significant (P<0.01), C, D, E, F no difference between groupsstatistically significant (P> 0.05).④C, D, E, F SOD activity in lung tissue of mice than the A, B group was significantly lower, the difference was statistically significant (P<0.01), D, E SOD activity lower than group C, There were significant differences (P<0.01), F group SOD activity is lower than C group, but the difference was not statistically significant (P> 0.05).⑤C, D, E, F mice lung tissue GSH-PX activity lower than A, B group, the difference was statistically significant (P<0.01), D, E, F mice lung tissue GSH-PX activity lower than Cgroup, the difference was statistically significant (P< 0.01/P<0.05).⑥C, D, E, F MDA content in lung tissue of mice than the A, B were significantly increased, the difference was statistically significant (P<0.01), D, E MDA content in lung tissue of mice was significantly higher than C,There was significant difference (P<0.01), F group MDA content increased than the C group, the difference was statistically significant (P< 0.05).⑦lung tissue biopsy can be observed in the A, B group of ciliated epithelial cells and complete rules of bronchial lumen, surrounded by no infiltration of inflammatory cells to pseudostratified ciliated columnar epithelial cells were predominant.C group, see off epithelial cells, infiltration of inflammatory cells around the wall, basement membrane thickening. D,E,F group,see off a larger number of epithelial cells, around the wall were a larger number of inflammatory cell infiltration, peripheral vascular edema.Conclusion:Inhalation of naphthalene may aggravate asthmatic reaction in mice.
