Aim:The study was undertaken to investigate the effect of recombinant growth hormone in the change of IL-6 on intestinal mucosa in rats with obstructive jaundice.Methods:Sixty adult male Wastar Rats were randomly divided into 3 groups:I, Sham Operation (SO, n=20);â…¡, Obstructive Jaundice (OJ, n=20);â…¢, Recombinant Human Growth Hormone (rhGH, n=20). In each group, rats were randomly assigned into 7days (n=10) and 14days (n=10) group. Group I:20 rats underwent laparotomy, but common bile duct (CBD) was not ligated and transected. Groupâ…¡andâ…¢:40 rats underwent laparotomy, and CBD was ligated and transected. In the group of rhGH, Rats were undertaken subcutaneous injected of rhGH on posterior limb wall each day with dosage 0.75iu/kg/day. On postoperative 7days and 14days, the blood of portal vein and the terminal segments of ileum were harvested. The TB,DB,ALT and endotoxin was measured by portal blood. Ileum stained with hematoxylin and eosin was examined histologically and morphometrically. The IL-6 of ileum was examined by immunohistochemistry and identified by Image Pro Plus (IPP).Result:(1) TB,DB,ALT in the groups of OJ and rhGH were average higher than those in the groups of SO (P<0.05); TB,DB,ALT in the groups of OJ and rhGH were gradually increased while the time going (P>0.05); There were not significant difference between the groups of OJ and rhGH on TB,DB,ALT, (P>0.05). (2) The ileal mucosa of group OJ and rhGH were acutely injuried than group SO (P<0.01).While the time going the degree of ileal mucosa injuried in group OJ was increased(P<0.05). The degree of ileal mucosa injuried in group rhGH-7days was lower than in group OJ-7days(P<0.05); The degree of ileal mucosa injuried in group rhGH-14days was lower than in group OJ-14days(P<0.01). The degree of ileal mucosa injuried in group rhGH-14days was lower than in group rhGH-7days, but there were not significant difference (P>0.05). (3) The level of endotoxin in the groups of OJ and rhGH were higher than those in the groups of SO (P<0.05); There were gradually increased as the time going (P<0.05); The level of endotoxin in the group of rhGH were less than those in the group of OJ (P<0.05). (4) The level of IL-6 in the groups of OJ and rhGH were less than those in the group of SO (P<0.01); The level of IL-6 in the groups of rhGH were higher than in the group of OJ (P<0.01); The level of IL-6 in the group of OJ-14days was less than those in the group of OJ-7days (P<0.05); There were not significant difference between the groups of rhGH-14days and rhGH-7days on the level of IL-6 (P>0.05).Conclusion:(1) Ectogenic growth hormone can improve the IL-6 secretion of intestine mucosal in rats with obstructive jaundice. It also can keep intestine mucosal structural integrated and function lower the level of endotoxin in portal blood.
