OBJECTIVE: The present study is to establish the rapid separation of components tanshinone method,and a measurement to test the content of 4 kinds of tanshinone in salvia, in order to improve the quality evaluation of Radix Salvia.METHOD: Based on the objective of the present study, we first collect the relevant literature of salvia.On this basis, we target to start a systematic experimental study. Salvia silica gel column chromatography Separation of fat-soluble extract, re-use high-speed counter-current chromatography separation and purification.The structures of the target compounds were identified by ESI-MS and NMR.Then we adopt a multi-assessment method to measure the quality evaluation of Radix Salvia together with experimental study. We use high-performance liquid to conduct a series of study, including a linear study of the 4 tanshinone, chromatography system precision study, stability of the test solution, method precision study, recoveries visits, tanshinoneⅡA on tanshinoneⅠ, tanshinone Cryptotanshinone and the relative correction factor dihydro reproducibility study, peak alignment parameters tested components investigated. Studies by the above method, TanshinoneⅡA built in as standard, simultaneous determination of Salvia miltiorrhiza 4 tanshinone ingredients of a more measured assessment method. 4 of 10 batches of Radix Salvia Tanshinone species composition are determined and measured respectively, using a multi-assessment method and external standard method to determine the content of tanshinoneⅡA, tanshinoneⅠand dihydro Cryptotanshinone tanshinone. We compare the results with external standard method evaluation with a multi-method test results.RESULT: With the method of Silica gel column chromatography with high-speed countercurrent chromatography,we successfully isolate tanshinone I,dihydro tanshinone I,tanshinoneⅡA ,,dihydro Tan,Trijuganone B and six monmer Cryptotanshinone,and conduct structural identification. Were measured using a multi-assessment method and external standard method for the determination of the 10 batches of Radix Salvia tanshinoneⅡA, tanshinoneⅠ,and dihydro Cryptotanshinone tanshinone content test showed that no significant difference between the two.CONCLUSION: The research work and the results obtained,will further improve the quality evaluation of Salvia miltiorrhiza level,and improve the quality standards of Salvia miltiorrhiza reference.
