| Objective In order to evaluate the quality of Salvia miltiorrhiza Bge. Better, study on the HPLC fingerprint ofliposoluble components and the protection effect of hypoxia on cardiac myocytes injured by oxygen/glucose deprivation(OGD),to establish spectrum-effect correlating mathematical modle.To provide a reference for scientific evaluation of the quality of Salvia miltiorrhiza Bge.Methods Salvia miltiorrhiza Bge. samples were collected from the whole nation,to investigate the extration methods of liposoluble components samples and chromatographic conditions,to ensure the internal standard, to establish HPLC fingerprint of liposoluble components;the cardiac myocytes are cultured in vitro,for establishment of OGD,the cardiomyocytes were assigned into control groupã€OGD group and liposoluble components group,to observe cell morphological changes and to determine the content of LDH in cell culture supernatant; analyse the results of HPLC fingerprint and the pharmacodynamic test,then established the spectrum-effect related mathematical modle by generalized regression neural network(GRNN).Results Established the methods of HPLC fingerprintof liposoluble components in Salvia miltiorrhiza Bge.,selected 14 peaks in common,established the methods of pharmacodynamics test.Got the resultsof 50 batch samples,the mathematical modle of spectrum-effeciency of liposoluble components in Salvia miltiorrhiza Bge. had been established by GRNN and verified.Conclusion The method of HPLC fingerprint measuring has good repeatability,the modle of spectrum-effect can predict pharmacological function by entering HPLC fingerprint date. |
