ObjectiveTo investigate the expressions of Interleukin-1αand Matrix metalloproteinase-13 in middle ear cholesteatoma epithelium of human.Then to analyze their corelations and roles in the hyperplasia and ossein destructiveness of human middle ear cholesteatoma epithelium.Methods1.37 cases of cholesteatoma specimens were collected at time of surgery, in the meantime,12 cases of the normal skin of external acoustic meatus were used as the control group. The samples were fixed in 10% Formalin liquor and made into paraffin section.2. According to the extent of ossein destructiveness,37 cases were divided into four groups:Ⅰdegree:5cases,Ⅱdegree:11 cases,Ⅲdegree:15cases,Ⅳdegree:6cases.3. Immunohistochemical staining was used to detect the expression of MMP-13 and IL-1αin the cholesteatoma and skin of external acoustic meatus. The computer image quantitative analysis was used to examin the expression of MMP-13 and IL-1α. 4. All data were analyzed statiscally by software SPSS 10.0.Result1. The expression of MMP-13 in cholesteatoma and skin of external acoustic meatus:The positive immunostaining of MMP-13 appeared all the epithelial layer of the cholesteatoma localized in the cytoplasm, appeared brown granular distribution,and it showed a tendency of gradual increasing from basal layer to cornrous layer, while there was no immunostaining for MMP-13 in normal skin of eternal acoustic meatus.2. The expression of IL-1αin cholesteatoma and skin of external acoustic meatus.The positive immunostaining of IL-la appeared all the epithelial layer of the cholesteatoma, and mail spread on basal layer, stratum spinosum. while there was no immunostaining for IL-la in normal skin of eternal acoustic meatus.3. The immunostaining for IL-la and MMP-13 were stronger as well as the degree of ossein destructiveness was stronger in human middle ear cholesteatoma epithelium.The was statistical significance between the groups. (P<0.05)4. The was statistical significance between the level of IL-la and MMP-13 in middle ear cholesteatoma epithelium.(r=0.452,P<0.05)Conclusion1. The expression of IL-1αand MMP-13 highly in human middle ear cholesteatoma epithelium,suggested that maybe they play an important role in the multiplication of the cholesteatoma epithelium.2. There was a direct ratio between IL-la,MMP-13 and ability of erosion of cholesteatoma,there may be a closely relation between IL-lα,MMP-13 and ability of erosion of cholesteatoma.3. The expression of IL-la and MMP-13 correlated to each other, indicated that they may have been synergistic action in multiplication and erosion of human middle ear cholesteatoma.
