Taohe Chengqi Decoction To Treating Chronic Non-bacterial Prostatitis And Effect Mechanism Of Optimization Formulations

Purpose:By the side of the subject in the combined experience of leptin on side (Liu Siphonostegia, wild chrysanthemum, litchi) in the treatment of Chronic Abacterial Prostatitis (CAP) clinic on the basis of good results obtained by animal experiment of leptin Tang decoction side experience, granules, capsules three formulations of CAP in rats, the initial exploration of the prescription treatment of the disease mechanism; comparing three different formulations of the herbs on the efficacy of CAP in rats, To explore the combined experience of leptin on the best side formulations.Method:3-month-old male wistar rats were 72, were randomly divided into 2 groups:60 in model, such as by Zhang Yaqiang rat model of autoimmune CAP modeling; saline group ether anesthesia,12 were given equal amount of physiological intraperitoneal injection of saline and multi-point intradermal injection.45 days, in the model with the saline group 12 rats were randomly removed, orbital blood samples to detect TNF-α, to determine the success of modeling. Saline group after successful only as a control group of 12 groupⅠ; 60 in the model were randomly divided into 5 groups: saline group after 12 successful only as a control group I group; 60 rats were randomly divided into pathological groupⅡ, the forefront of the Thai groupⅢand leptin on the experience side decoction combined groupⅣ,Ⅴgroup granule group, capsule groupⅥgroup,12 in each group, and appropriate drug treatment. After 4 weeks, abdominal blood serum levels of tumor necrosis factor (TNF-α) orally, then were killed, prostate tissue, the average prostate tissue, prostate wet weight/body weight ratio of wet weight detection; by immunohistochemistry the expression of iNOS in prostate tissue; and light and electron microscopy of the pathological changes of prostate tissue.Results:Compared with the control group, model group, the pathological prostate wet weight, wet weight/body weight ratio increased levels of serum TNF-α, iNOS expression of prostate tissue, significant difference (P<0.01), light microscopy, electron microscopy pathological Prostate tissue showed inflammatory changes, suggesting that the animal model of success.Nuclear decoction of three formulations of combined experience side reduced the average wet weight of prostate tissue, prostate wet weight/body weight ratio, and pathological model group (P<0.01, P <0.05), to good effect decoction. Leptin on the three formulations of combined experience side reduction of serum TNF-αlevels, compared with the pathological model (P<0.01), to good effect decoction; leptin on the three formulations of combined experience side reduced iNOS expression in prostate tissues, decoction obvious; light and electron microscopy showed leptin on the pathological side of three formulations of combined experience in both improving the role of inflammation in prostate tissue, is superior to decoction.ResultsExperimental study found that:(1) leptin on combined experience side decoction, capsules, granules of these three formulations have a good effect on the CAP, and the decoction of the best. (2) The combined experience of leptin on the mechanism of Treatment of CAP may be related to local immune regulation function of the prostate and reduce prostate inflammatory cytokines TNF-α, to reduce infiltration of inflammatory cells, inhibit proliferation of fibrous tissue and restoration of prostate secretory function.
