Objection:To study the effect and mechanism of Endostar inhibitor hela cell in proliferation and the capacity of invasion, discuss the mechanism associated with VEGF and MMP-9, to provide theoretical and experimental basis for the targeted therapy of cervical cancer.Methods:1,Hela cells were cultured and treated by Endostar with different concentration (1,5,10,100μg/ml) for 24,48and72 hours, the effect of Endostar on hela cell were analyzed by MTT assay, respectively, and calculate the inhibition rate.2, Endostar treated hela cells(1,10,100μg/ml) degree for 24 hours and their migration ability were observed by wound assay.3,Hela cells treated by Endostar with different concentration (1,10,100μg /ml) for 24 hours, and then RNA was extracted. The expression of VEGF, MMP-9 mRNA Was measured by RT-PCR.Results:1,MTT shows that different Endostar on hela cell after 24,48and72 hours, Cell growth was inhibited to varying degrees, on dose dependence. Each experimental group and control group were significantly different (p<0.05).2,24 hours after the scratch, Control cells are basically covered the original scratch, the experimental group(1,10,100μg/ml)with increasing concentration of endoststin,Hela cells migrated to the scratch area gradually reduced, as compared with the control group, the width did not significantly change.3. RT-PCR showed the experimental group(1,10,100μg/ml)VEGF mRNA expression decreased with the concentration gradually increased,the gray value(0.151±0.165,0.066±0.150,0.002±0.012)was significantly smaller in the control group(0.340±0.184).MMP-9 mRNA expression also decreased with the concentration gradually increased,the gray value(0.759±0.006,0.561±0.007, 0.198±0.025)was also significantly smaller in the control group(1.335±0.043). Each experimental group and negative control group,the expression differences had statistically significant.(P<0.01)Conclusion:Endostar inhibited the proliferation of Hela cell in a dose time-dependent manner. Endostar can inhibit the migration of Hela cell. Endostar can down-regulate mRNA expression of VEGF and MMP-9 in Hela cells. The mechanism may be associated with the inhibit of the tumor metastasis factor on the expression of VEGF, MMP-9 expression to inhibit cervical cancer invasion and metastasis.
