Objective:To observe the influence of Xuezhikang on expression of ICAM-1,VCAM-1 in culture HUVECs induced by insulin (INS), angiotensinⅡ(AngⅡ), to investigate the insulin and AngⅡ-induced atherosclerosis and the non-lilid mechanism of Xuezhikang reductase inhibitors on anti-atherosclerosis.Methods:Human umbilical vein endothelial cells strain (HUVECs) was cultured in vitro. Passage 2-3 of HUVECs were used for experiment,then divided into eight groups, the control group; Xuezhikang (150μg/ml); INS (100nmol/ml); Angll ( 5×10-8mol/ml); INS+Angll; INS+Xuezhikang; Angll+Xuezhikang, INS+AngⅡ+Xuezhikang. Cells in the groups containing Xuezhikang were incubated with Xuezhikang for two hours before co-incubated with INS or/and Angll. The HUVECs were coincubated with INS, AngII,Xuezhikang for 24,48,72h, and then observing cell morphology and cell proliferation by microscope and MTT assay.The level of ICAM-1,VCAM-1 mRNA was measured by reverse transcription-polymerase chain reaction (RT-PCR) and in the same time the protein of ICAM-1 and VCAM-1 was measured by western blotting.Results:Xuezhikang is not toxic to the endothelial cells. INS, Angll make endothelial cell body longer, cytoplasmic density, nuclear condensation and reduce the number of cells,and have a synergistic effect.Compared with the control groups,the levels of ICAM-1 and VCAM-1 mRNA and protein in the INS or/and Angll groups are higher,once INS and Angll are combined, the levels of ICAM-1 and VCAM-1 mRNA and protein are obviously higher than that when either is employed lone. Xuezhikang could inhibit the expression of ICAM-1 and VCAM-1 that induced by INS and AngⅡ. the control and Xuezhikang groups are not obviously different.Conclusions: Both insulin and AngⅡcan change cell morphology,decrease cell proliferation and promote endotherlial cells to express ICAM-1 and VCAM-1, and meanwhile have a synergistic effect. Xuezhikang can inhibit the expression of ICAM-1 and VCAM-1. The inhibitory effect of Xuezhikang maybe independent on cholesterol-lowing effect.
