Experimental Study Of Acorus Gramineus Soland And Polygala Powder On Acute Toxicity Testing

Objective:To investigate acute toxicity reaction of Acorus gramineus soland and Polygala powder (Acorus gramineus soland 5g, Polygala 5g) in mice. To appraise median lethal dose and 95%confidence interval of Acorus gramineus soland and Polygala powder. The results will provide experiment foundation for subac and chronicity toxicity reaction.Methods:The weight 22±2g of healthy female Kunming mice were selected and were randomly labeling with picric acid. A "Up-down" method of acute toxicity testing was applied. The initial oral dose was 175mg/kg, the follow-up dose was given in accordance with the dose AOT425 irrigation software.The general state and abnormal toxicity state were observed. Moreover, LD50 and 95%confidence interval were calculated. To evaluate noxious property by globally harmonized classification system and EU toxicity grading standards.Results:1. The general state:The experiment mice of intragastric administration with Acorus gramineus soland and Polygala powder, Acorus gramineus soland extract and Polygala extract were normal in weight, eating and drinking water.2. Abnormal toxicity symption:â‘ action:The action is decreasing when the dose of Acorus gramineus soland and Polygala powder and Polygala extract exceeded 1750mg/kg.â‘¡reaction of nervous system:The tremble appeared when the dose of Acorus gramineus soland and Polygala powder and Polygala extract exceeded 5000mg/kg.â‘¢reaction of automatic nervous system:All mice were no abnormalities.â‘£death:The mice died when the dose of Acorus gramineus soland and Polygala powder exceeded 5000mg/kg. The seven mice died five when the dose exceeded 5000mg/kg in Acorus gramineus soland extract and Polygala extract.3. LD50å'Œ95%CI:LD50 is 3129mg/kg and 95%CI is 1750-5000mg/kg in Acorus gramineus soland and Polygala powder; LD50 is 5000mg/kg and 95%CI is 2615-6480mg/kg in Acorus gramineus soland aqua extract. LD50 is 1750mg/kg and 95%CI is 756-3400mg/kg in Polygala aqua extract.4. Acorus gramineus soland and Polygala powder is five category by GHS and U grade by EUTGS. Acorus gramineus soland aqua extract is not elsewhere classified by GHS and Ugrade by EUTGS. Polygala aqua extract is four category by GHS and harmul by EUTGS.Conclusions:1.
