Objective:By detecting PTEN, PI3K.p110a, AktSer473 and Ki-67 protein expression in ovarian serous carcinoma (OSC) to analysis the mechanism of OSC, provides a reference diagnostic molecular markers.Methods:(1) Selected 42 cases ovarian serous carcinoma(OSC) in which ovarian high grade serous carcinoma (HGSC)35 cases, ovarian low grade serous carcinoma (LGSC)7 cases, ovarian clear cell carcinoma (CCC)7 cases, ovarian serous borderline tumors (OSBT)18 cases, ovarian serous cystadenoma (OSA) 10 cases, normal ovarian epithelial tissues (NOT)5 cases, were paraffin-embedded tissue.(2) Each group ovarian serous tumors (OST) were detected PTEN, PI3K.p110a, AktSer473 and ki-67 protein expression with immunohistochemical SP method and analysis the correlation of protein expression as well as among various groups of the protein in the OSC (3) Analysis the correlation between protein expression and the histological grade, invasion and lymph node metastasis in OSC.Results:(1) PTEN protein-negative rate (68%) higher than OSBT (22.0%) in the OSC, OSA (30.0%) and NOT (0%) group,χ2=17.046, P<0.01; between OSC and OSBT,χ2= 10.481, P <0.01; between OSC and NOT, P<0.05; Analysis linear trend of PTEN protein expression in OST,P<0.01;between LGSC and HGSC the P= 0.066 is close to 0.05.(2) PI3K.p110a positive rate (95%) higher than OSBT (89.0%) of OSC, OSA (10.0%) and NOT (0%) groups,χ2= 46.839, P<0.01; between OSC and OSBT,χ2= 0.057, P> 0.05; both of between OSC and OSC,between OSA and NOT,P<0.01; analysis linear trend PI3K.p110a protein expression in OST,P<0.01.(3) AktSer473 showed the positive rate (74%) higher than OSBT (56.0%) in OSC, between OSA (10.0%) and NOT (0%) groupsχ2= 19.649, P<0.01; between OSC and OSBT,χ2= 1.840, P> 0.05; between OSC and OSA, between OSC and NOT both P<0.01; analysis of linear trend between OST and AktSer473,P<0.01.(4) Among PTEN,PI3Kp110αand AktSer473 expression was a negative correlation in OSC (r=-0.347; r=-0.494, P<0.05); correlation of the expression between PI3K.p110a and AktSer473 was positive (r= 0.394, P <0.05). (5)Ki-67 protein expression in OSC than OSBT, OSA, and NOT (F= 49.894, P<0.01); OSC and CCC between the F=-4.916, P<0.01; LGSC and HGSC between the F=-4.591, P< 0.01; between the two groups Ki-67 protein expression and Akt protein positive expression in OST (F= 40.180, P<0.01).Conclusions:(1) The absence of PTEN protein expression involved in the onset and progress of OSC, PTEN protein loss rate from NOT to the OSB,OSA then OSC were increased, the highest in the HGSC, there may be differences between the HGSC and LGSC groups.(2) PI3Kp110a and AktSer473 protein expression, involved in the occurrence and progression of OSBT and OSC, but also from the NOT to OSA and OSBT then OSC,were increased.(3) Lack of PTEN expression and PI3Kp110αand AktSer473 protein positive expression in the occurrence and progress of the OSC may have a synergistic effect. (4) Ki-67 proliferation index is the highest in the HGSC,and NOT is the lowest, there are differences of the groups between HGSC and LGSC, suggesting that activity of the proliferation is highest in the HGSC. |
