ObjectiveTo study the effects of Yangjing Zhongyu decoction on androgen synthesis for the prenatally androgenized female rats.MethodsPregnant rats were treated with daily subcutaneous injections of testosterone propionate from embryonic days 16 to 19, and female offspring were studied as subject .The rats were treated with Yangjing Zhongyu decoction compound particle for 30 days, which of estrogen cycle was observed,and the level of estradiol( E2), testosterone(T),as well as 17a-hydroxyprogestrone(17-OHP) were measured by radioimmunoassay.The changes of ovarian morphological changes were detected by Hematoxylin and Eosin (HE) Staining and the cellular expression of key signal molecules were analyzed by RT - PCR technology.Results1.The prenatally androgenized female rats were ideal model.2. Overy histological anatomy shows that there are more atretic follicles and polyfolliculars and less corepus luteums and granular layers in model group than in control group.3.Compared with C group,there is an increase( P<0.01) of serum 17-OHP and T in M group , whereas that of E2 is on decline( P<0 . 01 ). While in the Y3 group , all the above-mentioned abnormalities were retorted to some extent(P<0. 01 or P<0. 05) as compared with those in the M group.4. Compared with M group, there is an increase of FSHR, IGF-1 and StAR mRNA in 3 YJZYD groups,especially in Y3 group( P<0.01).Conclusion1. The prenatally androgenized female rats were ideal model with PCOS.2. Yangjing Zhongyu decoction compound particls can reduce androgen, promote the ovulation and improve ovarian function as well as regulative reproductiv of PCOS model animals.3. Yangjing Zhongyu decoction can up-regulate expressions of FSHR, IGF-1 and StAR mRNA in ovary, regulate ovarian function and promote follicular development in PNA rat .
