【Objective】To investigate estrogen receptor gene polymorphisms and depressive disorder in Han nationality menopausal women of Guangzhou.【Methods】The Pvu-II , Xba-I polymorphisms of estrogen receptorαgene and the Rsa-I ,Alu-I polymorphisms of estrogen receptorβgene were analyzed in 62 patients with depressive disorder and 72 menopause healthy controls by PCR-RFLP method.【Result】1.The age,education years, rate of Progesterone and Testosterone were not significantly different between the two groups(P>0.05)2.the rate of HAMA,HAMD and E2 were significantly different between the two groups(P<0.01).3.The outcome of ERαgene polymorphisms analysis3.1 The genotype distributions(PP,Pp and pp) were significantly different between the two groups(P<0.05);The genotype PP was significantly more in the depression group than in controls; and allele frequencies(P and p) of Pvu-II polymorphisms were not significantly different between the two groups(P>0.05); Compared with wild—type pp.the susceptibility of precocious puberty with genotypes PP and Pp Was respectively 5.95(95%CI:1.50-23.54)and 1.46(95%CI:0.70—3.02)fold。3.2 The genotype distributions(XX,Xx and xx) were significantly different between the two groups(P<0.05);The genotypeXx was significantly more in the depression group than in controls; and allele frequencies(X and x) of Xba-I polymorphisms were not significantly different between the two groups(P>0.05); Compared with wild—type xx.the susceptibility of precocious puberty with genotypesXX and Xx Was respectively 1.14(95%CI:0.29-4.45)and 2.46(95%CI:1.19—5.09)fold.4. The outcome of ERβgene polymorphisms analysis4.1 The genotype distributions(rr,RRandRr) and allele frequencies(R and r) of Rsa-I polymorphisms in exon 5 were not significantly different between the two groups(P>0.05);4.2 The genotype distributions(aa,AAandAa)of Alu-I polymorphisms in exon 8 were not significantly different between the two groups(P>0.05);however, the allele frequencies(A and a) of Alu-I polymorphisms in exon 8 were not significantly different between the two groups(P>0.05);5. As a reference to PP-type ,the the level of E2 in Pp gemetype wsa higher than PP(OR=1.013);and the rate of HAMD,HAMA were all lower than PP(OR=0.959and 0.968);As a reference to PP-type,the level of E2 in pp genetype was higher than PP(OR=1.012), and the rate of HAMA and MoCA were all higher than PP(OR=1.496 and 1.067),however, the rate of HAMD was lower than PP genetype(OR=0.647). As a reference to Xx-type, the rate of MoCA in XX genetype was lower than Xx(OR=1.419); As a reference to Xx-type the rate of HAMD in xx genetype was lower than Xx(OR=-0.262),however, the rate of HAMA was higher than Xx genetype (OR=0.277). 【conclusion】1. Decreased estrogen levels associated with the onset of depression2. The patients with depression disorder have barrier in Cognitive function3. The polymorphisms of Pvu-II of ERa gene had relation to depression disorder in menopause women and the mutant gene increased depression morbidity risk.women with PP genotype were the most susceptible to depression disorder.The polymorphisms of Xba-I of ERa gene had relation to depression disorder in menopause women and the mutant gene increased depression morbidity risk.women with Xx genotype were the most susceptible to depression disorder.4. The Rsa-I polymorphism in exon 5 of estrogen receptorβgene was not associated with the risk getting the depressive disorder in Guangzhou .The Alu-I polymorphisms in exon 8 of estrogen receptorβgene was not associated with the depressive disorder in Han nationality menopausal women of Guangzhou .5.The incidence of depression disorder in PP gene-type was related with low estrogen,and was affected in cognitive.the Xx genetype were at high risk incidence in depression disorder,but was not related with low estrogen...
