The effect of different (3-cyclodextrin derivatives (hydroxypropyl-(3-cyclodextrin, sulfobutyl ether-β-cyclodextrin, randomly-methyl-β-cyclodextrin) on aqueous solubility and permeability through semi-permeable cellophane membrane (MWCO 500) of capsaicin (CP) was investigated. Furthermore, we investigated CP solubility and permeation of ternary systems with hydroxypropyl-β-cyclodextrin (HPpCD) in the presence of water-soluble polymers (PVP and HPMC).In the solubility studies, both binary and ternary systems showed an AL type of phase solubility diagram, the solubility and the stability constant (Ks) of binary systems were in the order:RMβCD> SBEβCD> HPβCD, and the ternary systems showed higher solubility and stability constant (Ks) of CP than CP-HPPCD binary system, and the the stability constant (Ks) showed an increase with the increased concentration of water-soluble polymer both for PVP and HPMC.In the permeation studies, for the binary systems, the permeation flux (Js) were in the order:HPβCD> SBEβCD> RMβCD, which is inverse to the stability constant (Ks) of CP/CD binary systems; and for the ternary systems, both PVP and HPMC showed a decrease of the permeation flux (Js) of CP than the CP-HPβCD binary system, The permeation flux (Js) of CP dereseased with the increasing of PVP concentration and increased with the increasing of HPMC concentration.This work demonstrates that there exists a negative correlation between the stability constant (Ks) and permeation flux (Js) of drug in drug/CD binary systems; for the ternary systems, the stability constant (Ks) and permeation flux (Js) of CP-HPβCD-PVP ternary systems showed a negative correlation, but the the CP-HPβCD-PIPMC ternary systems showed a positive correlation of stability constant (Ks) and permeation flux (Js).
